罗氏沼虾促雄性腺提取物对性别的影响及一种促雄性腺激素类似物的RNA干涉

摘 要: 将罗氏沼虾(Macrobrachium rosenbergii)的促雄性腺提取物及原核表达的促雄性腺激素类似物(rMAL),通过心脏注射的方式注射到卵巢刚发育的雌性沼虾体内。结果显示:注射促雄性腺提取物的雌虾卵巢发育明显减慢,卵巢中多膜状结构,无卵母细胞存在。注射原核表达促雄性腺激素类似物的雌虾卵巢发育没有明显变化。同时,还将体外合成的促雄性腺激素类似物双链RNA,注射到性发育成熟的雄虾体内进行RNA干扰。结果表明:壶腹中目的基因表达水平明显下降,但未观察到雄性生殖系统的外观表型变化。建立了对促雄性腺提取物及其类似物的生物学活性分析和RNA干扰的有效方法,为进一步开展后续的研究打下坚实的基础。

Title: Effect of androgenic gland extract on sexual characters and RNAi of an androgenic gland hormone like protein in the giant freshwater prawn, Macrobrachium rosenbergii

Abstract: In this study, androgenic gland extract and recombinant androgenic gland hormone like protein (rMAL) of the giant freshwater prawn, Macrobrachium rosenbergii, were injected to mature females. The ovary development of androgenic gland extract injected females was obviously delayed. There were a lot of membranes instead of oocytes in the ovaries. The females with injected rMAL had no change in ovary development. At the same time, the dsRNA of androgenic gland hormone like protein gene (MAL) was synthesized and injected to mature male shrimps. The injection caused a distinct decrease in target gene expression level in ampullae, but no exterior appearance change of male reproductive system was observed. In summary, the results from the protein biological analysis and RNA interference experiments had well demonstrated a feasible and convenient method for further study.
