MMP-9/TIMP-1在香烟暴露的大鼠肺组织中的表达

摘 要: 目的:探讨香烟暴露对慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)大鼠模型肺组织及人血管内皮细胞MMP-9,TIMP-1表达的影响。方法:1) SD雄性大鼠24只,随机分为正常对照组(N组)和COPD模型组(M组),采取烟熏法建立COPD模型。观察2组大鼠一般指标、体重、肺功能、肺组织病理学变化及支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)中白细胞计数和分类,肺组织中MMP-9,TIMP-1的表达变化。2) 应用免疫印迹(Western blot)检测不同浓度香烟提取物(cigarette smoke extract,CSE)作用人血管内皮细胞MMP-9,TIMP-1表达变化趋势。结果:1) M组大鼠出现毛发脱落、反应迟钝、呼吸频率加快等表现,N组大鼠与实验前相比无明显变化。2) N组大鼠体重、肺功能指标、炎性细胞计数、肺组织形态学指标、肺组织MMP-9,TIMP-1,MMP-9/TIMP-1表达与M组比较,差异有统计学意义(P<0.05)。3) 随着CSE作用浓度增高,人血管内皮细胞MMP-9,TIMP-1表达逐渐增强。结论:1) 吸烟是COPD的一个重要致病因素;2) 肺内MMP-9/TIMP-1的失衡在COPD的发生发展中可能起重要作用;3) 血管内皮细胞MMP-9和TIMP-1可能在COPD发生中起一定作用。

Title: Expression of matrix metalloproteinase-9 and tissue inhibitor of metalloproteinase-1 in rat lung tissue after cigarette-smoking exposure

Abstract: Objective: To study the expressions of matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of matrix metalloproteinase-1 (TIMP-1) in the lung tissue of rat with chronic obstructive pulmonary disease (COPD) and vascular endothelial cell after cigarette-smoking exposure. Methods: Twenty-four male SD rats were randomly divided into a normal control (N) group and a COPD model (M) group. The rat model of COPD was established by exposure to cigarette smoking. Rats’ general condition, weight changes, lung function, pathologic features of lung tissues and inflammatory cell differentials in bronchoalveolar lavage fluid (BALF) and expression intensity of MMP-9 and TIMP-1 in lung tissue were investigated. The vascular endothelial cell was stimulated by different concentrations of cigarette smok extract (CSE) and Western blot was carried out to examine the expression of MMP-9 and TIMP-1. Results: There were some changes such as falling off of hair, dullness and breathing in M group’s rats while the rats of group N had no evident change. There were significant differences in the body weight, lung function index, inflammatory cell count, lung histomorphologic index and expressions of MMP-9 and TIPM-1 between groups N and M. The expressions of MMP-9 and TIMP-1 in the vascular endothelial cells increased with the concentration of CSE. Conclusion: Smoking is one of key etiological factors for COPD. The imbalance of MMP-9/TIMP-1 in lung tissue and the proteolytic enzyme produced by vascular endothelial cell may play an important role in the pathogenesis of COPD.
