亚砷酸钠亚慢性染毒对小鼠肝和脑组织抗氧化水平的影响

摘 要: 目的:探讨亚砷酸钠(NaAsO2)亚慢性暴露对小鼠肝脏和大脑组织脂质过氧化水平及抗氧化作用的影响。方法: 采用亚慢性毒性实验方法。昆明种小鼠60只,按体重随机分为正常对照组(C)、低剂量组(L)、中剂量组(M)和高剂量组(H)4组,分别自由饮用蒸馏水、0.6,2.5,10.0 mg/L的亚砷酸钠水溶液,连续染毒90 d,实验结束后测定小鼠肝脏和大脑组织中脂质过氧化产物丙二醛(MDA)的含量,超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和谷胱甘肽S-转移酶(GST)的活力。结果:随着亚砷酸钠染毒剂量的增加,小鼠肝脏和大脑组织中MDA含量增加(P<0.05),SOD,GSH-Px活力降低(P<0.05);肝脏和大脑组织中GST活力随着砷染毒剂量的增加呈现先升高后降低的趋势(P<0.05)。结论:亚砷酸钠亚慢性染毒可使小鼠肝脏和大脑组织脂质过氧化水平增强,抗氧化能力降低,且氧化/抗氧化水平的改变存在组织特异性。提示脂质过氧化可能是砷毒性作用的机制之一。

Title: Influence of subchronic sodium arsenite poisoning on antioxidation level of mouse liver and brain tissues

Abstract: Objective: To investigate the influence of subchronic sodium arsenite(NaAsO2) exposure on lipid peroxidation and antioxidation in mouse liver and brain tissues. Methods: Subchronic toxicity experiment was applied. 60 Kunming mice were randomly divided into four groups according to their body weights, including control group (C), low dose arsenic poisoning group (L), middle dose arsenic poisoning group (M), and high dose arsenic poisoning group (H). Mice of the control group were fed with distilled water. Mice in group L, group M and group H were respectively fed with water with dissolved sodium arsenite at concentrations of 0.6 mg/L, 2.5 mg/L and 10.0mg/L. The experiment lasted for 90 d. Then the content of lipid peroxidation product malondialdehyde (MDA) and activities of the total superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and glutathione S-transferase (GST) in the mouse liver and brain tissues were determined respectively. Results: With the increase of the sodium arsenite dose, the MDA content significantly increased (P<0.05), and the activities of SOD and GSH-Px significantly decreased (P<0.05); and with the increase of the sodium arsenite dose, the GST activity increased firstly and decreased subsequently (P<0.05). Conclusion: The subchronic sodium arsenite poisoning can increase lipid peroxidation and decrease antioxidation in mouse liver and brain, and the oxidation/antioxidation level variation has tissue specificity. It indicates that lipid peroxidation may be one of the mechanisms of liver and brain injury in arsenic poisoning.
