噬菌体-指示菌模型体外评价抗病毒药物的初步研究

摘 要: 建立一种利用噬菌体-指示菌模型体外评价抗病毒药物的新方法。采用此实验模型对抗病毒药物双黄连、病毒唑和甘草酸单铵进行了噬菌体抑制实验。为了排除实验药物对模型系统的干扰,首先研究了药物对指示菌(大肠杆菌)的毒性作用,实验结果表明:双黄连和病毒唑对指示菌有一定的毒性作用,具有剂量依赖性,而甘草酸单铵无此作用。噬菌体-指示菌模型测定3种药物药效的实验结果表明:双黄连和病毒唑具有明显抑制噬菌体作用,双黄连和病毒唑分别与噬菌体作用20 h后的抑制活性最强,抑制率分别为89�69%和47�50%,而甘草酸单铵则未表现出对噬菌体的抑制作用。这些实验结果建议噬菌体-指示菌模型可用于体外评价抗病毒药物,具有安全、快速、简便易行的特点,适于从大量天然产物中初筛抗病毒药物。

Title: Preliminary research on in vitro evaluation of antiviral drug by phage-indicator model

Abstract: This paper develops a novel method which evaluates antiviral drug in vitro by phage-indicator model. The phage inactivation experiments were carried out with antiviral drugs Shuang Huanglian (SH), Ribavirin and Monoammonium Glycyrrizinate (MG). Firstly the toxicity of 3 drugs to indicator (E. coli) was determined to get rid of interference of the drugs in the model system. The results indicated that SH and Ribavirin had toxicity to the indicator, which was relative to their doses. The drug act of 3 drugs was detected in the model. The data demonstrated that the phage survival was significantly inhibited by SH and Ribavirin. After the interaction of the drugs and phage for 20 h, the high inhibition rate of SH and Ribavirin to phage survival reached to 89.69% and 47.50% respectively, while MG had no inhibition to phage. On the base of these experimental results, evaluation of antiviral drugs in vitro by means of the phage-indicator model is feasible and practicable, which is suitable for screening of antiviral drugs, especially for those isolated from a large number of natural products.
