马鹿-牛种间核移植胚胎的克隆和培养

摘 要: 应用体细胞核移植技术,将马鹿皮肤成纤维细胞与牛去核卵母细胞融合,构建马鹿异种体细胞核移植胚胎。实验结果表明:马鹿成纤维细胞可以在牛去核卵母细胞内去分化,构建的马鹿-牛体细胞异种核移植胚胎可以发育到囊胚阶段,囊胚率为15�1%. 同时构建了牛同种体细胞核移植胚胎作为对照组,马鹿异种核移植胚胎卵裂率与对照组相比差异不显著,但是囊胚发育率显著(P<0�05)低于对照组(15�1% vs 22�5%),Hochest 33342荧光染色表明异种核移植的囊胚细胞数显著(P<0�05)低于对照组(68�4个vs 83�8个)。

Title: In vitro culture of wild red deer (Cervus elaphus)-bovine heterogenerous embryos generated by cloning

Abstract: Red deer fibroblast cells were fused into enucleated bovine oocytes, and red deer-bovine heterogenerous embryos were reconstructed by cloning. The experiment results showed that the reconstructed heterogenerous embryos could develop into blastocyst stage with the blastocyst rate 15.1%. Bovine homogenerous cloning embryos were reconstructed as the control group. The cleavage rate of the heterogenerous embryos were not significantly different with the control group, however, the blastocyst rate were significantly (P<0.05) lower than that of the control group(15.1% vs 22.5%), and the Hochest 33342 immunostaining showed that the cell number of the heterogenerous embryos were significantly (P<0.05)lower than that of the control group (68.4 vs 83.8).
