水稻籽粒硒含量的QTL定位及遗传分析

摘 要: 硒是人体不可缺少的微量元素,籽粒硒含量的数量性状位点(quantitative trait loci,QTL)定位和遗传分析对研究富硒水稻的遗传育种具有重要意义。以籼稻亲本奉新红米和明恢100杂交的一个145个株系的F2群体为实验材料,利用92个SSR(simple sequence repeat)标记对水稻硒含量进行了QTL定位研究。共检测到6个水稻籽粒硒的QTLs,位于第2,3,5,6,7染色体上,对表型变异的贡献率在4�10%~12�74%之间,其中,位于第3染色体的分子标记rm7和rm251之间的qSe3对表型变异的贡献率最大,为12�74%,其增效等基因来自亲本奉新红米,表现为显性效应。6个QTLs的联合贡献率为42�19%,具有基因累加效应。这有利于深入理解水稻硒含量的遗传基础,为硒含量的QTL精细定位和分子标记辅助选择奠定基础。

Abstract: Selenium (Se) has been recognized as an essential trace element for human beings and plays a number of functions important to human health. Understanding the genetic of Se contents in rice and quantitative trait loci (QTL) mapping are the basis for effective selection in Se breeding program. In the present study, a set of 145 F2 lines derived from the cross between Fengxin red rice and Minghui 100 were used to detect QTLs associated with Se contents with 92 SSR markers covering the whole rice genome. A total of 6 QTLs were detected for Se contents on chromosome 2, 3, 5, 6, 7, respectively. Accounting for general contributions range of 4.10%~12.74% to Se contents. qSe3 was located between rm7 and rm251 on chromosome3 with the largest general contributions 12.74% to phenotypic variation. The favorable allele of qSe3 for Se contents originated from Fengxin red rice and was controlled by dominant effect. And the total general contributions of 6 QTLs was 42.19% with cumulative effect. This helps to further understand the genetic basis for Se contents and will provide the basis for gene mapping of Se contents and molecular marker-assisted selection of Se in rice.
