摘 要: Islet1(ISL1)属于LIM homeodomain转录因子家族中的一员,近年来研究发现,ISL1在心脏发育的最早期阶段起关键性作用。isl1基因敲除会导致小鼠胚胎完全缺失流出道、右心室以及大部分心房,最终胚胎于10.5(E10.5) d死亡。谱系示踪分析表明,ISL1主要标记第二生心区(second heart field, SHF)的心脏前体细胞,对于这些前体细胞的增殖、迁移和存活必不可少。目前,已经从发育的胚胎、出生后胚胎的心脏,甚至基因特异性打靶的胚胎干细胞中分离出ISL1阳性前体细胞,这些细胞显示出巨大的多向分化潜能,不仅能分化为有功能的心肌细胞,而且还能分化为心脏所有其他类型的细胞。因此,ISL1阳性细胞代表了组织特异性心脏前体细胞,在再生医学和心肌损伤修复方面具有非常重要的治疗价值。

Title: The functions of ISL1, a LIM-homeodomain transcription factor in cardiac development

Abstract: ISL1 is a member of LIM-Homeodomain transcription factor family. Recent studies show that ISL1 plays a pivotal role for the earliest stage of heart development. isl1 knockout in mice leads to a completely missing of the outflow tract, right ventricle, and much of the atria, ultimately results in the mice to die at embryonic day 10.5 (E10.5). Lineage tracing analysis demonstrates that ISL1 mainly marks cardiac progenitors derived from the second heart field. It is required for proliferation, migration and survival of these progenitors. Currently, ISL1-positive progenitors have been isolated from embryonic heart, postnatal heart, and even gene specific-targeting embryonic stem cells. These cells exhibited robust multiple differentiation potential, which not only differentiated into functional cadiomyocytes, but also differentiated into all the other diverse cardiac cell types. Therefore, ISL1-positive cells represent tissue-specific cardiac progenitors, and have significantly valuable therapeutic effects in myocardial regeneration and injury. This paper focuses on the research advances of ISL1 in cardiac development during recent years.
