艾灸预处理对急性胃黏膜损伤大鼠胃黏膜细胞Cyt-C,Apaf-1及其细胞凋亡调控因子的影响

摘 要: 目的:通过观察艾灸预处理对急性胃黏膜损伤大鼠胃黏膜细胞色素C(cytochrome C,Cyt-C)、凋亡活化因子1(activator 1,Apaf-1)及其凋亡相关因子的影响,探讨艾灸诱导热休克蛋白70(heat shock protein 70,HSP70),通过线粒体信号转导途径抑制细胞凋亡,保护胃黏膜损伤的机制。方法:将32只健康Wister大鼠随机分为4组,即捆绑组(A)、模型组(B)、艾灸穴位组(C)、艾灸非穴组(D)。采用无水乙醇灌胃法制备大鼠急性胃黏膜损伤模型。检测各组Cyt-C,HSP70,胃黏膜细胞凋亡指数(apoptotic index,AI),Apaf-1,Caspase-9和Caspase-3的表达。结果:与A组相比,B组HSP70, AI,Cyt-C,Apaf-1,Caspase-9和Caspase-3含量明显增加(P<0.01或P<0.05);与B组相比,C组HSP70表达明显增加(P<0.01),而AI,Cyt-C,Apaf-1,Caspase-9,Caspase-3均显著降低(P<0.01);与C组相比,D组大鼠HSP70表达显著降低, 而AI, Cyt-C,Apaf-1,Caspase-9,Caspase-3显著升高(P<0.05)。结论:艾灸通过上调大鼠胃黏膜细胞HSP70表达,影响细胞凋亡线粒体信号转导途径相关因子,抑制胃黏膜细胞凋亡,达到保护胃黏膜损伤的作用。

Title: The effect of pre-moxibustion on Cyt-C, Apaf-1 and gastric mucosal cell apoptosis regulation factors in the acute gastric mucosa injury rats

Abstract: Objective: To observe the effect of pre-moxibustion on cytochrome C (Cyt-C), apoptosis activator 1(Apaf-1) and apoptosis-related factors of gastric mucosal cells of module rats, and to explore the mechanism of protecting gastric mucosal damage by heat shock protein 70(HSP70) induced by pre-moxibustion. Methods: 32 healthy rats were randomly assigned into 4 groups, namely control, model, moxibustion at acupoints and moxibustion at non-acupoints group with 8 cases in each group. Rat models of acute gastric mucosal damage were established by gastric injection of absolute ethyl alcohol in all the groups except the control group. The expression of cytochrome C (Cyt-C), expression of HSP70, cell apoptosis index (AI), apoptosis activator 1 (Apaf-1), Caspase-9 and Caspase-3 in gastric mucosa were detected. Results: Compared with control group, HSP70, AI, Cyt-C, Apaf-1, Caspase-9, Caspase-3 in model group were significantly increased (P<0.01 or P<0.05). Compared with model group, HSP70 was significantly increased (P<0.01),but AI, Cyt-C, Apaf-1, Caspase-9, Caspase-3 were significantly decreased in moxibustion at acupoints group (P<0.01). Compared with moxibustion at acupoints group, HSP70 was significantly decreased, but AI, Cyt-C, Apaf-1, Caspase-9, Caspase-3 were significantly increased in moxibustion at non-acupoints group (P<0.05). Conclusion: Moxibustion can up-regulate the expression of HSP70 in gastric mucosa, and then act on targets related to apoptosis mitochondrial signal transduction pathway, to inhibit gastric mucosal cell apoptosis and protect gastric mucosa. And the protecting effect has a certain specificity of acupoints.
