高压氧、地塞米松对大鼠超长随意皮瓣FGF-1及TGFβ-1表达量的影响

摘 要: 目的 探讨高压氧、常压氧对大鼠随意皮瓣成活率的影响。方法:Wister雄性大鼠60只,随机分为三组,每组于背部中线形成一蒂在尾端的超长随意皮瓣,A组不给于其他干预方式;B组术后即刻给予地塞米松5mg皮瓣下注射;C组术后当天开始每天给予高压氧2次,每次1 h,持续5天,第7天统一处理动物,计算皮瓣成活率。结果 术后7天测量各组大鼠平均成活率分别为:对照组:(48.6±8.0)%;地塞米松组成活率(81±4.4)%;高压氧组:(89.1±3.7)%;组间差异明显,p<0.05。高压氧组和地塞米松组较对照组VEGF及CD105表达明显提高,但高压氧组与地塞米松组之间没有差异性,而TGFβ-1则三组均为阴性或弱阳性,组间没有差异性。结论 高压氧与地塞米松均可以促进随意皮瓣的成活,但促进成活的机理可能存在差异。

Title: Research of the effect of hyperbaric oxygenation and normalbaric oxygenation on the survival rate of random-pattern flaps in rats

Abstract: Objective To study the effect of hyperbaric oxygenation and normalbaric oxygenation on the survival rate of random-pattern flaps in rats. [Methods] : Forty-five Wister rats were assigned to 3 groups equal in number randomly. A free flap was elevated in the central part of the rachis. Group A was left as controls. In group B, all the rats received normalbaric oxygenation twice a day until postoperative day 5. In group C, hyperbaric oxygenation was applied to the rats twice a day lasting for five days. On the seventh day, survival rates for all flaps were calculated. Results On the seventh day, the survival rates were as follows: control team,(48.6±8.0)%; normal baric oxygenation, (51.7±7.1)%; hyperbaric oxygenation team, (89.1±3.7)%.The difference was significant among the teams. Expression level of FGF-1 was much higher in the hyperbaric oxygenation team than the left two. And there is no significant difference among the three teams on the TGFβ-1 expression. Conclusion Hyperbaric oxygenation and dexamethasone used in this study significantly increase flap survival rate in rat random-pattern flap model, while the mechanism might be different.
