二氢丹参酮Ⅰ对宫颈癌Hela细胞的放射增敏作用及对细胞周期的影响

摘 要: 目的:研究二氢丹参酮Ⅰ(dihydrotanshinoneⅠ)对人宫颈癌细胞株Hela的放射增敏作用及对细胞周期的影响。方法:四氮唑噻唑兰/四甲基偶氮唑盐(MTT)法检测不同浓度二氢丹参酮Ⅰ分别在24,48,72 h对Hela细胞的抑制作用;克隆形成实验分析,多靶单击拟合模型方程计算放射增敏比,评价增敏效果。流式细胞仪检测二氢丹参酮Ⅰ对Hela细胞周期和早期凋亡的影响。结果:不同浓度(0.5~16.0 mg/L)二氢丹参酮Ⅰ对Hela细胞的毒性均有明显剂量和时间依赖性,其24,48,72 h的IC50值分别为8.39,2.18,1.48 mg/L.二氢丹参酮Ⅰ对Hela细胞具有放射增敏作用,细胞克隆形成法得到其增敏比为1.35. 二氢丹参酮Ⅰ可使Hela细胞细胞周期的构成发生明显变化并诱导细胞凋亡。结论:二氢丹参酮Ⅰ对宫颈癌细胞系Hela有较好的细胞毒性和放射增敏作用,其增敏机制可能使Hela细胞生长停滞在G0/G1期并诱导细胞凋亡,使S期细胞比例降低,从而加强射线对肿瘤细胞的杀伤能力。

Title: The effect of dihydrotanshinone I on radiosensitization effects and cell cycle of Hela cells of human cervical cancer

Abstract: Objective: To investigate the radiosensitization effects and cell cycle of dihydrotanshinoneⅠon Hela cells. Methods: The MTT assay was used to detect the growth inhibition rates of Hela cells at 24, 48, 72 h which cultured with dihydrotanshinone I in different concentrations. With multi-target single-hit model, the radiosensitization was investigated by clone formation assay through sensitizing enhancement ratio (SER). The cell cycle distribution and apoptosis were measured by flow cytometry. Results: The different concentrations (0.5~16.0 mg/L) of dihydrotanshinoneⅠhad cytotoxicity on Hela cells, which were clearly dose and time-dependent. The IC50 of 24, 48, 72 h were 8.39, 2.18, 1.48 mg/L, respectively. The radiosensitization of dihydrotanshinoneⅠwas proved significantly on Hela cells. Further clone formation assay indicated that the sensitization enhancement ratio was 1.35. Cells were treated with dihydrotanshinone Ⅰwhich had significant effects on cell cycle of Hela cells, and induced apoptosis. Conclusion: Dihydrotanshinone I has significant cytotoic and radiosensitization effects on human cervical cancer. One of the mechanisms may be that it may make significant G0/G1 phase arrest and induced apoptosis, and decrease in S phase, thus cells are more sensitive to radiation.
