表皮生长因子受体及基质金属蛋白酶-2在食管癌的表达特点及意义

摘 要: 目的:探讨食管癌组织中表皮生长因子受体(epidermal growth factor receptor,EGFR)和基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)的表达关系。方法:应用免疫组织化学及蛋白印迹实验检测66例人食管癌组织及瘤旁正常组织EGFR及MMP-2的表达,评价EGFR及MMP-2在患者性别、年龄、肿瘤TNM(tumor,lymph node,metastasis)分期、病理分级等临床参数组的表达。Pearson相关性检验分析EGFR及MMP-2表达相关性。结果:免疫组化显示EGFR,MMP-2在肿瘤组织表达的平均灰度净值分别为25.42±3.15,37.46±6.18,在瘤旁正常食管组织平均灰度净值分别为5.03±3.49,9.80±2.24,组织间表达水平相比均有显著的统计学差异(P<0.01)。免疫组化及蛋白印迹表达分析显示EGFR,MMP-2的表达分布在TNM分期、病理分级和淋巴结转移中均有统计学差异(P<0.05)。免疫组化检测两蛋白表达净灰度值相关系数R=0.425,P=0.038;蛋白印迹检测2种蛋白表达相关系数R=0.674,P<0.05.结论:EGFR及MMP-2在食管癌均存在过表达,且二者的表达呈正相关,显示EGFR及MMP-2与食管癌发生、发展密切相关,为食管癌多靶点联合治疗提供依据。

Title: Expression and correlation of epidermal growth factor receptor and matrix metalloproteinase-2 in human esophageal carcinoma

Abstract: Objective: To detect the expression and relation of epidermal growth factor receptor (EGFR) and matrix metalloproteinase-2 (MMP-2) in human esophageal carcinoma. Methods: In 66 human esophageal carcinoma tissues, the expressions of EGFR and MMP-2 were detected by immunohistochemistry and Western-blot. The expressions of EGFR and MMP-2 in various clinicopathologic analyzed. Correlation analysis between MMP-2 and EGFR was analyzed by Pearson correlation coefficient. Results: By immunohistochemistry, the average gray scale values of EGFR in esophageal carcinoma and normal tissues were 25.42±3.15, 5.03±3.49, which showed the significant difference (P<0.01). And the average gray scale values of MMP-2 in esophageal carcinoma and normal tissues were 37.46±6.18 and 9.80±2.24, which showed a significant difference (P<0.01). The significant differences were observed in TNM-stage, lymph node metastasis and tumor classification in both the expression of EGFR (P<0.05) and MMP-2 (P<0.05) by immunohistochemistry and Western-blot. Correlation analysis between EGFR and MMP-2 by immunohistochemistry (R=0.425, P=0.038) and Western-blot respectively(R=0.674, P<0.05) suggested a good positive correlation. Conclusion: The data suggests that EGFR and MMP-2 overexpress in human esophageal carcinoma. Combination of EGFR and MMP-2 could be the useful markers for tumorigenesis and differentiation, which provides basis for therapy of human esophageal carcinoma.
