ABT-418对ADHD动物模型SHR自主活动的影响

摘 要: 为探讨烟碱受体激动剂烟碱异恶唑衍生物[(S)-3-methyl-5-(1-methyl-2-pyrrdidinyl)isoxazde,ABT-418]对注意缺陷多动障碍(attention deficit and hyperactivity disorder,ADHD)动物模型自发性高血压大鼠(spontaneously hypertensive rat,SHR)自主活动水平的影响,以哌醋甲酯为对照,采用开场实验对用药后SHR在4个不同时点的自主活动情况进行检测,并采用免疫组化方法检测皮层和海马的α4,β2烟碱受体表达水平。结果显示:ABT-418能减少SHR在开场实验中的爬格子数;ABT-418组的皮层α4,β2受体亚型、海马的α4受体亚型表达显著增强(P<0.05),而海马的β2受体亚型的表达水平没有显著改变。说明ABT-418能通过nAChRs的介导降低SHR的自主活动水平,连续使用后可导致nAChRs受体的向上调节。

Title: The effect of ABT-418 on the locomotor activity in SHR: an animal model of ADHD

Abstract: To investigate the effect of ABT-418 on the locomotor activity in spontaneously hypertensive rat (SHR), this study selected methylphenidate as control, and performed open field test after injection, half an hour and 24 hours later and 7th and 14th day. The results suggested that injection of ABT-418 in SHR can reduce the grating in open-field test. Compared with other groups, ABT-418 group was significantly higher in the protein expression of α4 and β2 in both cortex and hippocampus, yet the expressions of β2 in hippocampus were no significant difference (P<0.05). The result of the experiment indicated that ABT-418 may exert its effects through α4,β2 nicotinic receptor, and repeated administration of ABT-418 could induce the up-regulation of the nAChRs through certain mechanism.
