猪SGCB基因编码序列的克隆与序列分析

摘 要: 肌聚多糖-β基因(beta-sarcoglycan,SGCB)的缺失将会导致抗萎缩素蛋白复合体的消失,引起肌肉萎缩综合症2E型(limb-girdle muscular dystrophy type 2E,LGMD2E)。研究获得了猪SGCB基因的编码区序列。利用CLUSTALW,CDD,Signal P3.0,Prosite等软件对所编码蛋白质的结构、功能等特征进行了预测和分析。序列分析表明:该蛋白结构共有6个相关位点结合区域,其中包含2个铁硫结合区和凝血因子结合位点。系统进化树分析表明:该蛋白遵循动物进化规律,猪SGCB蛋白比人SGCB蛋白进化早。

Title: Cloning, sequencing and analysis of porcine SGCB gene

Abstract: Deficiency of beta-sarcoglycan (SGCB) will lead to limb-girdle muscular dystrophy type 2E (LGMD2E) due to vanish of anti-dystrophy protein complex. In this paper, coding region sequence of the porcine SGCB gene was obtained. CLUSTALW, CDD, Signal P3.0, and Prosite software was used to forecast and analyze the structure and function of the coded protein. Sequence analysis indicated that there exist 6 function domains, including 2 iron-sulfur binding region. The phylogenetic tree showed that the protein followed evolutionary law, and porcine SGCB protein was earlier than human's.
