N-Boc-Glu的合成方法优化

摘 要: 以甲醇作为均相反应介质,Boc酸酐与谷氨酸为原料,在碱性条件下,以产物熔点、羧基含量及产物纯度作为产物质量确定的指标,最终得出N-Boc-Glu的最优合成条件。通过实验小样确定反应时间为23 h左右;反应物料的物质的量配比(Glu∶Boc2O)为1.0∶1.5;反应温度为30℃;pH值为8~9;结晶条件是向溶液中加入乙酸乙酯(EtOAc)后再酸化的最优合成条件。

Title: Optimization of synthesis method of N-Boc-Glu

Abstract: The paper takes methanol as homogeneous reaction medium, Boc-anhydride and glutamic acid as material, and alkaling condition as reaction condition. The optimal synthesis condition was determined by measuring the melting point, the content of carboxylgroup and the product purity. The results show that the best experiment condition is as follows: The proportion of adding amount proportion of reactants (Glu∶Boc2O) is 1.0∶1.5, the temperature of the reaction is room temperature, the time of the reaction is about 23 hours, the pH value is 8-9, and the crystallization condition is the optimal synthesis condition of acidification after EtOAc is added to the solution.
