农杆菌介导SOS1基因转化烟草及转化植株抗盐性的初步检测

摘 要: 通过农杆菌介导法将盐敏感(salt overly sensitive,SOS)突变体SOS1基因转入珊西烟草中。对转化后T0代植株进行聚合酶链反应(polymerase chain reaction, PCR)检测,初步证明目的基因已经整合到烟草基因组中。对转基因T3代幼苗及其愈伤组织、细胞进行抗盐生理检测。研究结果证实,在盐胁迫下,转基因烟草比野生型(wild type,WT)表现出更高的耐盐性。

Title: Initial survey on Agrobacterium-mediated SOS1 gene transformation of tobacco and salt tolerance test of transgenic tobacco

Abstract: SOS1 gene, which is a salt overly sensitive (SOS) mutant, was transferred into tobacco by Agobacterium-mediated transformation. The polymerase chain reaction (PCR) analysis indicated that SOS1 gene had been inserted into tobacco genome in some T0 generation transgenic plants. The callus, cells, and seedlings were detected in some T3 generation transgenic plants, confirmed that transgenic tobacco had greater ability to tolerate salt stress compared to wild type (WT) plants.
