川芎嗪联合贝那普利对并发肾病GK大鼠肾组织sICAM-1的影响

摘 要: 目的:观察川芎嗪联合贝那普利对并发肾病GK(Goto-Kakizaki)大鼠肾组织sICAM-1表达水平的影响。方法:将40只雄性GK大鼠随机分为模型组、贝那普利组、川芎嗪组、川芎嗪+贝那普利组(即联合组);每组10只,分别灌服相应药物,共16周。实验第8,12,16周将大鼠放入代谢笼中,收集24 h尿液,测定24 h尿蛋白定量;治疗16周后,采用放射性免疫法(平衡法)测定TNF-α,双抗体夹心酶联免疫吸附测定(enzyme-linked immuno sorbent assay,ELISA)法测定IL-6,免疫组化法测定肾脏sICAM-1的表达。结果:实验结束时,各治疗组Scr,BUN,24 h尿蛋白定量,IL-6,sICAM-1较模型组均有显著下降(P<0.05或P<0.01),其中以联合组下降趋势最明显(P<0.01);各治疗组Ccr均有所增高(P<0.05);联合组还可明显降低血清TNF-α水平(P<0.01)。川芎嗪联合贝那普利能下调肾组织sICAM-1表达水平,降低血清Scr,BUN水平,提高内生肌酐清除率,减少尿白蛋白的排泄,降低TNF-α,IL-6水平,从而改善糖尿病肾病(diabetic nephropathy,DN)大鼠的肾功能。结论:川芎嗪联合贝那普利能改善DN大鼠肾功能,改善肾组织炎症病变。下调肾组织sICAM-1表达和降低TNF-α,IL-6水平可能为其作用机制之一。

Title: Effects of ligustrazine combined with benazepil on the expression level of soluble intercellular adhesion molecule-1 in GK rats’ nephridial tissue

Abstract: Objective: To observe the effects of ligustrazine combined with benazepil on the expression level of soluble intercellular adhesion molecule-1 in Goto-Kakizaki (GK) rats’ nephridial tissue with diabetic nephropathy. Methods: Forty GK male rats were randomly divided into four groups: model group, ligustrazine group, benazepil group and ligustrazine combined with benazepil group, 10 every groups. GK rats received corresponding medicines according to the experimental design for 16 weeks. The rats were put into metabolism cages in the 8th week, the 12th week and the 16th week, collecting urine to detect proteinuria in 24 h. After treating for 16 weeks, ICAM-1 was detected by immunohistochemistry, the level of TNF-α by radioimmunoassay and IL-6 by enzyme-linked immuno sorbent assay (ELISA) in blood serum. Results: At the end of experiment, the level of Scr, BUN, proteinuria in 24 h, TNF-α, IL-6, ICAM-1 were all decreased in all therapeutic groups compared with model group (P<0.05 or P<0.01), ligustrazine combined with benazepil group in particular (P<0.01). Ccr was increased in all therapeutic groups compared with model group (P<0.05). Ligustrazine combined with benazepil group can down regulated the expression of sICAM-1, decrease the level of Scr, BUN, TNF-α and IL-6, and reduce the excretion of proteinuria to improve renal function in GK rats with diabetic nephropathy. Conclusion: ligustrazine combined with benazepil can improve renal function in GK rats with diabetic nephropathy inflammation change of nephridial tissue. To degrade the express level of ICAM-1 and reduce the level of TNF-α, IL-6 is one of the possible mechanisms.
