HCMV编码蛋白pUL23对Hela细胞增殖和细胞周期的影响

摘 要: 目的:pUL23是人巨细胞病毒(human cytomegalovirus,HCMV)编码的蛋白,对病毒的繁殖具有调控作用,研究pUL23对Hela细胞增殖和细胞周期的影响,为阐述pUL23功能的分子机理提供依据。方法:以转染pcDNA和pcDNA-UL23-Flag的Hela细胞为模型,用噻唑蓝(MTT)比色法检测pUL23对细胞增殖的影响;用碘化丙啶(PI)染色法检测pUL23对细胞周期的影响;western blotting检测细胞周期蛋白表达水平的变化。结果:pUL23加速Hela细胞周期从G0/G1→S期过渡,但对细胞增殖无影响;与阴性对照相比,过表达pUL23时,细胞周期蛋白P21的表达下调,P16的表达水平不发生变化。结论:pUL23可能通过影响Hela细胞周期发挥调控病毒自身繁殖的作用。

Title: Effect of HCMV encoding protein pUL23 on Hela cell proliferation and cell cycle

Abstract: Objective: pUL23 is a protein that is encoded by human cytomegalovirus (HCMV) and it regulates propagation of the virus. This study aims to research into pUL23 on Hela cell proliferation and cell cycle, meanwhile provides the basis for demonstrating the molecular mechanism of pUL23 functions. Methods: Used the Hela cells with transfection of pcDNA and pcDNA-UL23-Flag as models, MTT assay was used to measure the impact of pUL23 on cell proliferation; pUL23 on cell cycle was detected by PI staining; cell cycle protein expression level changes were evaluated by western blotting detection. Results: The results showed that pUL23 accelerated the Hela cell cycle from G0/G1→S phase transition, but had no effect on cell proliferation; compared with the negative control, the expression of cell cycle protein P21 was significantly reduced and no significant difference was found in the expression level of P16 when pUL23 was over-expressed. Conclusion: It is concluded that pUL23 could regulate virus self-reproduction by affecting Hela cell cycle.
