棘白菌素B脱酰酶产生菌N-04-06菌株的发酵条件优化

摘 要: 采用响应面分析法对棘白菌素B(Echinocandins B, ECB)脱酰酶产生菌N-04-06菌株发酵条件进行优化。首先用田口设计确定影响显著的因素和水平范围,然后利用响应面对显著因素的最佳水平范围进行研究,确定影响ECB生产的主要因子及其最适取值:蔗糖14.3 g/L、蛋白胨12.8 g/L、棉籽饼粉15.3 g/L、培养温度28℃.以该优化工艺条件进行50 L发酵罐试验。结果表明:分段控制pH值、温度、通气量和搅拌转速以及进行适时的流加补充碳源,可使菌株生长繁殖能力强、产酶效果好,且菌体干重在56 h达到21.2 g/L,酶活在发酵72 h达到16.2 U/L.

Title: Optimization of fermentation conditions for Echinocandins B deacylase production by strain N-04-06

Abstract: In this paper, to study the effect of fermentation conditions on the production of Echinocandins B (ECB) deacylase by strain N-04-06, response surface method was used to optimize the fermentation conditions in this study. Taguchi design was used to examine the significance of factors, and to confirm the level of each factor. The optimum level ranges of the significant factors were studied by using response surface. The optimal culture condition was confirmed: sucrose 14.3 g/L, peptone 12.8 g/L, cotton-seed powder 15.3 g/L, cultural temperature 28℃. The optimal condition was applied to produce ECB deacylase in a 50 L fermenter. The experiment result showed that the following conditions were suitable during ECB deacylase production: the stepwise control of pH, culture temperature, aeration rate and agitation speed. Through the utilization of these operation techniques, strain N-04-06 grew well with a higher dried mycelia weight 21.2 g/L after 56 hours fermentation, and the production was obtained with the stable enzyme activity higher than 16.2 U/L after 72 hours fermentation.
