图像分析系统在测量细胞DNA含量中的比较

摘 要: 目的:利用激光扫描共聚焦显微镜(laser scanning confocal microscope,LSCM)和图像分析仪在原位测量肝细胞(核)内化学物质含量,比较2种方法各自的特点。方法:选5只成年健康雄性小鼠,每只小鼠按常规方法制作3,5,7,15 μm的肝组织切片,Feulgen染色,用TIGER细胞图像分析系统和LSCM在组织原位测算以一个完整的二倍体、四倍体和八倍体肝细胞(核)为单位的DNA含量。结果:组织原位测量二倍体、四倍体、八倍体的单个完整肝细胞(核)内的DNA含量比值接近2或4倍。结论:应用2种分析图像分析系统通过适当的抽样和计算,可以较准确地计算出完整细胞(核)体积中化学物质的总量。

Title: Comparison of image analysis systems in measurement of nuclear DNA contents

Abstract: Objective: Using laser scanning confocal microscope (LSCM) and image analyzer for in situ measuring the chemical substances in liver nuclei, and comparing the characteristics of the two methods. Methods: 3, 5, 7, 15 μm liver tissue slices were prepared respectively from 5 adult male mice, and then stained together with Feulgen stain. The DNA contents in volume unit of whole diploid or tetraploid or octoploid nuclei were in situ measured and calculated by TIGER cell image analysis system and LSCM, respectively. Results: The ratios of DNA contents in volume unit of whole diploid, tetraploid and octoploid nuclei were close to 2 or 4. Conclusion: By using these two image analysis systems, the total chemical substance contents in full cell volume can be more accurately computed through appropriate sampling and calculation.
