孕期接触细菌脂多糖对成年子代种属行为、探索性活动及焦虑行为的影响

摘 要: 目的:细菌脂多糖(lipopolysaccharides,LPS)引起的宫内感染与啮齿类动物的神经发育损伤相关,研究主要探讨母体孕期接触LPS对成年F1代仔鼠种属行为、探索性活动和焦虑行为的影响。方法:孕鼠于受孕第8~15天每天经腹腔注射给予8 μg/kg LPS,阴性对照组孕鼠给予等容量生理盐水(normal saline, NS)。所有动物自然分娩。分别于仔鼠出生后第2个月与第6个月,每窝随机取雌雄各1只仔鼠进行行为测试。结果:种属行为实验发现,母体接触LPS明显增加其2月龄雄性仔鼠食物挖掘量,而其6月龄雌性子代食物储藏量却显著降低。旷场实验结果显示,孕期接触LPS明显增加6月龄雌性仔鼠平均周边时间及跨格数,而明显缩短第一格的潜伏期。结论:母体孕期接触LPS能够改变成年雌性子代种属行为、探索性活动和焦虑行为,并具有年龄依赖性。

Title: Effects of prenatal LPS exposure on species-typical behavior and exploration and anxiety activities in mice at adulthood

Abstract: Objective: Lipopolysaccharide (LPS)-induced intrauterine infection has been associated with neuro-developmental injury in rodents. The purpose of the present study is to explore the effects of prenatal LPS exposure on species-typical behavior and exploration and anxiety activities in adult mice. Methods: The pregnant mice were intra-peritoneally (i.p.) injected with LPS (8 μg/kg) daily from gestational day (gd) 8 to gd 15. A battery of neurobehavioral tasks was performed in mice at ages of 2 and 6 months old. Results: Species-typical behavior examination showed that prenatal LPS exposure markedly increased weight burrowed in 2-month-old male offspring and significantly reduced weight hoarded in 6-month-old female offspring. Open field test showed that the number of squares crossed and peripheral time, markers of anxiety and exploration activities, were markedly increased in 6-month-old female mice following prenatal LPS exposure. Interestingly, latency to the first grid crossing was markedly decreased in 6-month-old female mice whose mothers were exposed to LPS during pregnancy. Conclusion: Prenatal LPS exposure impairs species-typical behaviors, exploration and anxiety activities of female mice at adulthood in an age-dependent manner.
