补肾调肝法对ROS模型小鼠INHb-ACT-FS系统的影响

摘 要: 目的:探讨补肾调肝方乌萸汤对卵巢不敏感综合征(resistant ovarian syndrome,ROS)小鼠卵巢抑制素B(inhibin-B,INHb)-激活素(activin,ACT)-卵泡抑素(follistatin,FS)系统的影响。方法:以小鼠卵透明带3(zona pellucida 3, ZP3)建立ROS模型。用乌萸汤药低、中、高剂量进行治疗,以戊酸雌二醇组、模型组为阳性对照,双抗体夹心酶联免疫吸附技术(Sandwitch ELISA)检测各组小鼠血清INHb,ACT,FS的表达。结果:模型对照组血清INHb,FS含量降低,ACT升高,与空白对照组比较有显著性差异(P<0.05);用药各组INHb,FS含量均升高,ACT降低,较模型组有显著性差异(P<0.05);乌萸汤高剂量组INHb,FS含量高于西药组,其差异无统计学意义(P>0.05);乌萸汤各剂量组ACT含量均高于西药对照组,其差异无统计学意义(P>0.05)。结论:乌萸汤可改善模型小鼠免疫性卵巢炎症状并改善卵巢功能,说明补肾调肝中药可以调节卵巢内部因子,从而改善卵巢生殖内分泌功能。

Title: Effects of tonifying kidney and regulating liver on expression of INHb-ACT-FS protein in blood serum of immune ROS mice

Abstract: Objective: To probe the effects of Wuyu Decoction (WYT), a Chinese compound recipe for tonifying kidney and regulating liver, on expression of inhibin-B (INHb)-activin (ACT)-follistatin (FS) protein in mice blood serum with immune resistant ovarian syndrome (ROS). Methods: BALB/c female mice model of ROS was created by multiple sites subcutaneous injection of zona pellucida 3 (ZP3) to mice, and treated with low, middle and high dosage WYT, to detect the expression of INHb-ACT-FS protein in blood serum by ELISA method. Results: Comparing with the normal group, the expression of INHb, FS decreased and the expression of ACT increased obviously in model group, statistically significant (P<0.05). Comparing with the model group, the expression of INHb, FS increased and the expression of ACT decreased obviously in medication group, statistically significant (P<0.05). The expression of INHb, FS increased in the mice of dosage WYT than in the estradiol valerate group statistical insignificant (P>0.05). Comparing with the group of estradiol valerate, the expression of ACT increased in blood serum in the mice of WYT, statistically insignificant (P>0.05). Conclusion: WYT can relieve ovarian immune inflammation and improve ovarian function. It proves that tonifying kidney and regulating liver can adjust internal factors of ovarian, thus controlling reproduction and endocrine function.
