鲍鱼腹足酸性多糖的分离纯化、组成分析及其体外抗肿瘤活性研究

摘 要: 对鲍鱼腹足肌酸性粘多糖(abalone acid polysaccharide,AAP)进行提取、分离纯化及基本组成分析,并研究其体外抗肿瘤活性。鲍鱼腹足肌经木瓜蛋白酶酶解提取、氯化十六烷基吡啶(CPC)沉淀、盐溶、乙醇沉淀、透析、冻干得到鲍鱼腹足肌粗多糖,经DEAE离子交换层析和S-300凝胶过滤层析后得到一种纯度较高的新型AAP. 经鉴定其平均分子量(molecular weight,MW)为56.2 ku;柱前衍生高效液相色谱法分析单糖组成,绝大部分为氨基半乳糖(GalN)、葡萄糖醛酸(GlcUA)、岩藻糖(Fuc)和半乳糖(Gal),其物质的量之比为2.14∶2.37∶2.97∶1,此外还均含少量鼠李糖(Rha)和氨基葡萄糖(GlcN);离子色谱法测定硫酸基含量为15.5%. 体外抗肿瘤试验表明,AAP能够显著抑制小鼠S180肉瘤细胞和人肝癌细胞HEP-G2的增殖。

Title: Isolation, chemical composition and antitumor activity in vitro of the acid polysaccharide isolated from Haliotis discus hannai

Abstract: A novel glycosaminoglycan-like abalone acid polysaccharide (AAP) from the pleopods of Haliotis discus hannai was purified by DEAE ion exchange chromatography followed with S-300 HR gel filtration chromatography. Chemical composition analysis indicated that AAP was composed of galactosamine, glucuronic acid, fucose, galactose with a ratio of 2.14∶2.37∶2.97∶1, the content of sulfate was 15.5%. The average molecular weight (MW) was 56.2 ku in a TSK G4000 column. The antitumor activity of the sulfated polysaccharide in vitro was detected by MTT assay. It was found that AAP had an obvious antitumor activity in a dose-dependent manner in vitro and the maximum inhibitory ratio of S180 and HEP-G2 were 52.5% (800 mg/L) and 48.9% (800 mg/L), respectively.
