EGFR活化参与胃癌SGC7901细胞对TRAIL耐药

摘 要: 胃癌细胞对肿瘤坏死因子相关凋亡诱导配体(tumor necrosis factor-related apoptosis-inducing ligand,TRAIL)耐药机制尚不明确。研究明确了表皮生长因子受体(epidermal growth factor receptor,EGFR)活化是否参与胃癌SGC7901细胞对TRAIL耐药。利用Western blotting检测蛋白的表达,流式细胞术检测细胞凋亡,瞬时转染small interfering RNA(siRNA)抑制EGFR的表达。结果发现:TRAIL作用SGC7901细胞后,可以检测到EGFR的磷酸化。用EGFR的siRNA瞬时转染SGC7901细胞48 h,EGFR表达明显下调。与阴性对照相比,在干涉EGFR的细胞中,TRAIL诱导EGFR磷酸化的水平明显下降,表明抑制EGFR表达最终提高了TRAIL诱导的细胞凋亡(P<0.05)。因此,EGFR活化是胃癌SGC7901细胞对TRAIL耐药的原因之一。

Title: EGFR activation is involved in TRAIL resistance in gastric cancer SGC7901 cells

Abstract: The resistant mechanism of gastric cancer cells to tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) is still unclear. The research is to identify whether the activation of epidermal growth factor receptor (EGFR) is involved in TRAIL resistance in gastric cancer SGC7901 cells. In this study, protein expression was detected by Western blotting and cell apoptosis was determined using flow cytometry. Transient transfection with small interfering RNA (siRNA) was used to inhibit the expression of EGFR. The results showed that treatment with TRAIL induced the phosphorylation of EGFR in SGC7901 cells. After SGC7901 cells were transiently transfected with EGFR siRNA for 48 h, the expression of EGFR was obviously decreased. Compared to non-silencing control, TRAIL-induced EGFR phosphorylation was downregulated in EGFR knockdown clones. The inhibition of EGFR expression finally increased TRAIL-induced apoptosis in SGC7901 cells (P<0.05). Therefore, the activation of EGFR was one of the reasons of TRAIL resistance in gastric cancer SGC7901 cells.
