蝎毒多肽干预慢性粒细胞白血病小鼠模型p53与ras表达

摘 要: 目的:观察蝎毒多肽(peptide extracts from scorpion venom,PESV)干预慢性粒细胞白血病(chronic myelocytic leukemia,CML)NOD/SCID小鼠模型体内p53,ras的表达水平。方法:首先将K562细胞注入经过铯-137源照射的NOD/SCID小鼠体内,建立CML-NOD/SCID小鼠模型;对实验小鼠随机分组,用ELISA法检测小鼠p53,ras表达的蛋白水平。结果:PESV能够提升p53表达,抑制ras过度表达,其抑制效果与PESV浓度具有相关性。结论:PESV通过干预CML中p53,ras的表达有效地阻断了CML传变进展。

Abstract: Objective: To investigate the effect of peptide extracts from scorpion venom (PESV) on the expression of p53, ras in chronic myelocytic leukemia (CML)-NOD/SCID mice. Methods: Firstly, K562 cells were injected into NOD/SCID mice irradiated by Cs-137 and the CML-NOD/SCID mode of mice were established. Then, the mice were randomly divided into different groups and the expression level of p53 and ras proteins in serum was checked by ELISA. Results: PESV could increase the expression level of p53 and inhibit the overexpression of ras, and the inhibitory effect was related to PESV concentration.Conclusion: PESV can inhibit CML progress through affecting the expression of p53 and ras in CML cells.
