生活污水处理厂各环节中细菌含量及群落结构变化

摘 要: 将核酸染料SYBRs Green I荧光染色与流式细胞术(flow cytometry, FCM)相结合,用于分析污水处理厂各主要环节中细菌的总量,并利用巢式PCR-变性梯度凝胶电泳(denaturing gradient gel electrophoresis, DGGE)技术对生活污水处理厂各环节的水样细菌群落结构进行分析。该水厂包括初沉池、活性污泥处理池及二沉池3个主要环节。由于活性污泥法中产生的污泥絮凝体影响细菌数量的准确定量,需要使用超声方法对样品进行前处理后再进行FCM法检测。FCM法检测结果表明:细菌浓度范围在3.91×106~2.24×109 counts/mL之间。原水总细菌浓度为1.74×108 counts/mL,初沉池为1.22×108 counts/mL,活性污泥阶段增至2.24×109 counts/mL,二沉池总细菌又下降至3.87×108 counts/mL,出水总细菌浓度为3.91×106 counts/mL. 巢式PCR-DGGE结果分析表明:原水与初沉池样品相似度最高,相似性指数可达83.3%,而相似度最小的为活性污泥池和二沉池样品,相似性指数仅为27.9%.

Title: Variation of total bacterial cell counts and bacterial communities in different water treatment processes from a wastewater treatment plant

Abstract: In this study, fluorescence staining of microbial cells with the nucleic acid stain SYBR Green I together with quantitative flow cytometry (FCM) were used to analyze bacterial concentrations in water samples from a wastewater treatment plant. The variation of bacterial structure during each treatment was also investigated by a nested PCR-denaturing gradient gel electrophoresis (PCR-DGGE) approach. The plant treated wastewater through three steps: primary settlement, activated sludge and a secondary settlement process. Due to the complex aggregation of bacteria in activated sludge flocs, the exact number of cells present in untreated activated sludge was not known and could not be calculated precisely. Therefore, ultrasonication was applied to disaggregate activated sludge flocs and obtain a suspension made up mostly of free single cells, suitable for FCM analysis. The results from FCM showed that in the wastewater treatment plant, bacterial abundance varied from 3.91�06 counts/mL to 2.24�09 counts/mL. Total bacterial counts were 1.74�08 counts/mL in the influent, 1.22�08 counts/mL in the primary settled water, and increased to around 2.24�09 counts/mL in activated sludge flocs. In the secondary settled water, the bacterial counts decreased to 3.87�08 counts/mL and finally lowered to 3.91�06 counts/mL in the effluent. Nested PCR-DGGE profiles demonstrated that bacterial communities in influent and primary settlement samples were grouped into one cluster with the highest similarity coefficient of 83.3%, and the lowest similarity coefficient of 27.9% was observed between activated sludge and the secondary settlement process.
