污染物对青海弧菌Q67时间依赖微板毒性分析

摘 要: 以青海弧菌Q67为受试生物,96孔板为暴露实验载体,通过优化菌液与长期培养基混合比例,在Q67生长周期内选取9个暴露时间节点(即0.25,2,4,6,8,12,16,20,24 h),建立污染物或其混合物在不同暴露时间内对Q67发光抑制毒性的微板测试方法——时间依赖微板毒性分析(time-dependent microplate toxicity analysis, T-MTA);应用T-MTA测定5种对Q67基本无急性毒性的抗生素,即硫酸安普霉素(apramycin sulfate,APR)、氯霉素(chloramphenicol,CHL)、硫酸巴龙霉素(paromomycin sulfate,PAR)、硫酸链霉素(streptomycin sulfate,STR)和盐酸四环素(tetracycline hydrochloride,TET)在不同时间内的发光抑制毒性。结果表明:这些抗生素对青海弧菌Q67的毒性具有明显时间依赖特征。

Abstract: Taking Vibrio qinghaiensis sp. Q67 (Q67) as test organism and 96-well microplate as exposure experiment carrier, we developed a time-dependent microplate toxicity analysis (T-MTA) by optimizing the ratio of bacterium suspension to long-term medium and setting nine time points, such as 0.25, 2, 4, 6, 8, 12, 16, 20, 24 h, in the growth cycle of Q67. The T-MTA was used to determine the time-dependent toxicities of five antibiotics, apramycin sulfate (APR), chloramphenicol (CHL), paromomycin sulfate (PAR), streptomycin sulfate (STR) and tetracycline hydrochloride (TET). The antibiotics do not basically produce the acute toxicities to Q67. However, the results of the time-dependent toxicities showed that the toxicities of antibiotics displayed the significant time-dependent features.
