利用表面等离子体共振研究TB-EPS在表面的吸附

摘 要: 为了解紧密结合型胞外聚合物(tightly bound extracellular polymeric substances, TB-EPS)在表面的吸附特性,精确测定TB-EPS在表面上的吸附速率和亲和力,研究从序批式生物膜反应器(sequencing biofilm batch reactor,SBBR)生物膜中提取TB-EPS,采用表面等离子体共振(surface plasmon resonance, SPR)研究TB-EPS在金表面上的吸附情况。结果表明:TB-EPS在金表面吸附反应的结合速率常数(ka)为(31.7±5.2) L/(g·min),解离速率常数(kd)为(0.405±0.060)min-1,平衡结合常数(KA)为(78.3±24.4)L/g,平衡解离常数(KD)为(0.013±0.004)g/L. 因此,采用SPR能够方便快速地得到TB-EPS在表面的吸附动力学过程和吸附亲和力。

Abstract: In order to understand the adsorption characteristic of tightly bound extracellular polymeric substances (TB-EPS) on surface, and determine the adsorption rate and affinity accurately, in this study, the adsorption behaviour of TB-EPS extracting from a sequencing batch biofilm reactor (SBBR) was investigated using surface plasmon resonance (SPR). The results showed that the parameters of the TB-EPS adsorption reaction on gold surface were the following: association rate constant (ka) =(31.7 �5.2) L/(g穖in), the dissociation rate constant (kd) =(0.405 �0.060) min-1, the equilibrium binding constant (KA) =(78.3 �24.4) L/g, and the equilibrium dissociation constant (KD) =(0.013 �0.004) g/L. Therefore, it is fast and conveniently for the SPR to obtain the adsorption kinetics and adsorption affinity of TB-EPS on the surface.
