石英砂负载过氧化物酶的制备及应用初探

摘 要: 以石英砂为载体,研究在其表面固定辣根过氧化物酶(horseradish peroxidase, HRP)的方法。石英砂表面用壳聚糖进行预处理,然后利用层层组装技术,通过静电吸引将酶层层负载到其表面。HRP的固定化回收率和活性回收率分别达到70%和78%. 固定化酶的最适pH值在4.5左右,比游离酶略有降低。固定化酶的热稳定性和储存稳定性较游离酶有明显提高。利用固定化酶可有效降解污染土壤中的硝基苯,显示出其在土壤污染修复方面的应用潜力。

Title: Preparation of horseradish peroxidase immobilization on quartz sands and its application

Abstract: In this paper, a method of immobilization of horseradish peroxidase (HRP) onto quartz sands was developed. The sands were pretreated with chitosan and the enzyme was loaded onto their surface thereafter via layer-by-layer assembly technology based on electrastatic interaction. The immobilization recovery and activity recovery of HRP was 70% and 78%, respectively. The optimal pH for the immobilized enzyme was 4.5, which was lower than free enzyme. The thermo stability and longevity of HRP were significantly improved after immobilization. It was found the immobilized HRP could effectively degrade nitrobenzene in soil, which demonstrated its potential application in soil remediation.
