聚乙二醇衍生物对氟尿苷-层状双金属氢氧化物纳米杂化物的表面修饰

摘 要: 层状双金属氢氧化物(layered double hydorxide,LDH)的表面修饰对其作为纳米药物载体时的分散性、稳定性及生物相容性具有重要意义。以氟尿苷(floxuridine,FL)插层LDH(FL-LDH)纳米杂化物为模型体系,以具伪装隐形作用的聚乙二醇衍生物聚乙二醇单甲醚癸二酸酯盐(poly(ethylene glycol) monomethyl ether-sebacate salt, MPS)为修饰剂,研究药物—LDH纳米杂化物的表面修饰作用,制备出MPS修饰的FL-LDH纳米杂化物(简记为FL-LDH-MPS),并进行表征。结果表明:表面修饰可明显改善纳米杂化物的分散稳定性和药物缓释性能。由于聚乙二醇的生物兼容性和体内循环的“隐形”功能,这种药物输运体系具有重要应用前景。

Abstract: Surface modification of the layered double hydroxide (LDH) was of much importance for its stability, dispersity and biocompatibility as drug-carriers. In this paper, floxuridine (FL) intercalated LDH (FL-LDH) was modified with poly(ethylene glycol) monomethyl ether-sebacate salt (MPS). The obtained FL-LDH-MPS nanocomposites showed enhanced stability, dispersity and controlled drug release property. The obtained FL-LDH-MPS nanocomposites could have advantages as drug delivery system with controlled-release due to the biocompatible and water-soluble PEG shells with “stealth” function upon in-travenous injection.
