僵蚕抗氧化多肽的初步分离及活性分析

摘 要: | 采用酶解法制备僵蚕(Bombyx batryticatus)中抗氧化肽,通过超滤进行初步分离,分析其抗氧化活性。以1,1-二苯基-2-苦味酰基苯肼(DPPH)清除率为指标,筛选出胰蛋白酶作为最适蛋白水解酶,酶解条件为温度50℃,pH=6.9,酶量0.31 g/L,底物浓度3.0 g/L,酶解时间3.67 h,对DPPH清除率为60%. 采用超滤技术获得分子量3 ku以下的小分子肽,其DPPH清除率为96%. 僵蚕蛋白水解液对DPPH有50%清除率时,底物浓度为2 g/L,其总还原力、抑制脂质体氧化能力与对照抗坏血酸相当。高温下处理2.5 h内其抗氧化活性几乎没有变化,反复冻融6次对其抗氧化活性也几乎没有影响,但在常温下随保存天数的增加其抗氧化活性减弱。 |

Abstract: | In our current study, the antioxidant peptide was produced from enzymatic hydrolysate of white muscardin silkworm and was preliminary separated through the ultrafiltration. The antioxidant activity was analyzed. 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging activity was used as the criterion for antioxidant activity. Peptides derived from trypsin hydrolysate exhibited the highest DPPH radical scavenging activity. Protein was enzymatically hydrolyzed at 50℃, pH=6.9, 0.31 g/L for enzyme concentration, 3.0 g/L for substrate concentration, 3.67 h for enzymatic hydrolysis time. DPPH clearance was 60%. After ultrafiltration to obtain the low-molecular weight peptide less than 3 ku, the DPPH scavenging activity arrived at 96%. The concentration for DPPH scavenging activity of 50% was 2 g/L, the total reductive activity and liposome oxidation inhibition was quite equal with ascorbic acid. High temperature treatment (boiling water bath) for 2.5 h as well as repeated freeze-thaw for 6 times, seemed not to affect the antioxi-dant activity. But the antioxidant activity decreased with the increasing storage days at room temperature. |
