JNK信号转导通路在AngⅡ诱导的血管内皮细胞衰老中的作用机制

摘 要: 目的:探讨血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)对人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)衰老的影响,以及c-Jun氨基末端激酶(c-Jun-NH2-terminalkinase,JNK)、核转录因子-κB (nuclear factor-κB,NF-κB)表达的变化。方法:制备RPMI1640培养液(含10-6 mol/L AngⅡ)培养HUVECs,通过β-半乳糖苷酶(β-galactosidase,β-gal)染色、流式细胞仪检测细胞衰老;Western-blotting测定JNK,磷酸化JNK(p-JNK),NF-κB的蛋白水平。结果:AngⅡ诱导组β-gal阳性细胞数与对照组相比增长了(81.09±6.72)%;流式细胞仪检测细胞周期停滞于G0-G1期;与对照组相比,AngⅡ诱导16,32,48 h,p-JNK蛋白表达水平显著增高(P<0.05),p-JNK水平于32 h达到最高峰(P<0.01);p-JNK表达呈持续增加时,NF-κB呈显著上升趋势。结论:AngⅡ诱导内皮细胞的衰老可能与JNK/NF-κB信号转导通路有关。

Title: Effect of JNK signaling transduction pathway in vascular endothelial cell senescence induced by AngⅡ

Abstract: Objective: To explore the effect of angiotensinⅡ(AngⅡ) on senescence of human umbilical vein endothelial cells (HUVECs) and the expression change of c-Jun-NH2-terminalkinase (JNK) and nuclear factor-κB (NF-κB). Methods: HUVECs were cultured in RPMI1640 medium including AngⅡ (10-6 mol/L). β-galactosidase (β-gal) staining and flow cytometry were used to identify cell aging status. By means of Western-blotting, the expression of JNK, p-JNK and NF-κB was observed. Results: The number of positive cells which were identified by β-gal staining was significantly higher in AngⅡ-induced cells (81.09±6.72)% than those in the control cells. Flow cytometry showed that the cell cycle was at G0-G1 stage. Compared with control group, p-JNK, NF-κB protein expression increased markedly (P<0.05) induced by AngⅡ for 16, 32, 48 h, and the expression level of p-JNK reached the peak at 32 h (P<0.01). Furthermore, when the expression of p-JNK increased, the NF-κB expression also increased significantly. Conclusion: There is a probability that activated JNK/NF-κB signaling transduction pathway is involved in the process of endothelial cell senescence induced by AngⅡ.
