乙酰甲喹纳米药物的制备及其体外释放研究

摘 要: 目的:开发适合于注射给药的乙酰甲喹纳米粒子(MEQ@MSN)药物,以克服其味苦、水溶性差等问题。方法:合成高比表面积、高比孔容的MCM-41型介孔二氧化硅纳米材料(mesoporous silica nanoparticular,MSN),并以此纳米载体制得粒径均匀的MEQ@MSN. 实验分别通过扫描电镜(scanning electronic microscopy,SEM)、透射电镜(transmission electron microscope,TEM)、氮气吸附-脱附实验、傅里叶红外光谱等手段对载药前后的纳米粒子结构进行比较研究。结果:乙酰甲喹(mequindox,MEQ)成功载入到介孔二氧化硅的孔洞中,并形成平均直径约为100 nm的分散球状粒子。体外研究显示,MEQ可以快速从该纳米粒子中释放,以便及时发挥药效。结论:MEQ@MSN有望作为注射制剂用于养殖动物腹泻的治疗。

Title: Study on synthesis and in vitro release of mequindox nanoparticles

Abstract: Objective: To overcome the shortcomings of bitterness and hydrophobicity, the mequindox nanoparticles (MEQ@MSN) is planned to be studied for injection. Methods: MEQ@MSN were prepared with synthesized MCM-41 type mesoporous silica nanoparticular (MSN) which had characteristics of high surface area, total pore volume and uniform mesoporous structure. Both structures of MSN and MEQ@MSN were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), nitrogen adsorption-desorption and Fourier transform infrared spectra. Results: Mequindox (MEQ) was successfully loaded into the pores of mesoporous silica, and the formed the dispersed spherical particles with an average diameter of about 100 nm. In vitro studies showed that mequindox could be quickly released from MEQ@MSN, which made sure of mequindox to start work immediately. Conclusion: The MEQ@MSN is expected to be used as an injection of mequindox for treating of sick aquaculture animal.
