拟南芥miR858对干旱胁迫响应的研究

摘 要: | miRNA(microRNA)参与并调控植物生长、发育及非生物胁迫,包括盐(NaCl)胁迫、干旱胁迫等。利用RT-PCR方法,分析200 μmol/L 脱落酸(abscisic acid,ABA)、250 mmol/L NaCl干旱(脱水)处理拟南芥(Arabidopsis thaliana)3周龄苗后,miR858 初级转录本(pri-miR858)和miR858 成熟片段(miR858)的表达水平。结果表明:pri-miR858和miR858在ABA处理2 h后被诱导表达,pri-miR858于8 h达到峰值,16 h表达量下降;而miR858在8 h和16 h表达量逐渐上升。pri-miR858和miR858 在NaCl胁迫处理2 h后被诱导表达,在8 h和16 h表达量逐渐增高。pri-miR858和miR858 在干旱胁迫处理6 h后被诱导表达到峰值,12 h表达量下降。以上分析表明:miR858可以被ABA、盐、干旱胁迫诱导表达。研究构建了35S::miR858a载体,并转化拟南芥得到转基因株系,发现35S::miR858a植株更加耐旱。 |

Abstract: | miRNA (microRNA) plays important role in plant growth, development and responses to abiotic stresses including salt and drought. In this research, the expression of primary miR858 (pri-miR858) and mature miR858 (miR858) was analyzed using RT-PCR with 3-week-old Arabidopsis thaliana under the stresses of 200 μmol/L abscisic acid (ABA), 250 mmol/L NaCl and drought. Our analysis shows that the expression of pri-miR858 and miR858 are induced 2 h after treatment with ABA. Expression of pri-miR858 peaks at 8 h, and decreass at 16 h, while miR858 getting higher and higher at 8 h and 16 h. Both pri-miR858 and miR858 are induced 2 h after NaCl treatment and getting higher and higher at 8 h and 16 h. pri-miR858 and miR858 are induced 6 h after drought stress and decrease at 12 h. These results indicate that miR858 can be induced by ABA, NaCl and drought stresses. Moreover, miR858a over-expression vector is constructed and transformed to A. thaliana. Transgenic lines are obtained. We find that miR858 over-expression transgenic lines show drought tolerance. |
