金黄色葡萄球菌Pfk-2的克隆、表达、纯化、结晶及初步晶体学研究

摘 要: 磷酸果糖激酶-2(phosphofructokinase-2,Pfk-2)属于核糖激酶家族,由它催化的反应是整个糖酵解过程中的一个限速步骤,因此其在金黄色葡萄球菌(Staphylococcus aureus)的能量代谢中起到了十分关键的作用。为进一步了解Pfk-2的功能,首先表达并纯化得到了Pfk-2的蛋白质,通过座滴法筛选得到了Pfk-2与其底物果糖-6-磷酸(fructose-6-phosphate,F6P)的晶体,收集了X-射线衍射数据并进行了初步晶体学分析。Pfk-2及其底物复合物结构的解析对于研究其在金黄色葡萄球菌能量代谢过程中的作用有着关键性的意义。

Title: Cloning, expression, purification, crystallization and preliminary crystallographic analysis of Staphylococcus aureus Pfk-2

Abstract: Phosphofructokinase-2 (Pfk-2), which belongs to ribokinase family, catalyzes the key step in glycolysis and is a critical enzyme in the Staphylococcus aureus metabolism. In order to clarify its function, Pfk-2 was expressed and purified. Then the crystals of Pfk-2 and its substrate fructose-6-phosphate (F6P) were obtained using sessile drop method. And finally, the X-ray diffraction data of Pfk-2 crystal was collected and preliminary crystallographic analysis was carried out. The crystal structure of Pfk-2 and F6P has crucial significance for the study of its role in the energy metabolism of S. aureu.
