抗原呈递基质刚性对B细胞钙信号的影响

摘 要: 利用聚二甲基硅氧烷(poly-dimethylsiloxane,PDMS)材料模拟具有不同刚性的基质进行抗原呈递,通过激光共聚焦显微镜对B细胞识别呈递于不同刚性基质表面抗原后的胞内钙信号进行检测。结果表明,相比于识别低刚性基质表面呈递的抗原B细胞,识别呈递于高刚性基质表面的抗原B细胞钙振荡信号频率更高,并且振动幅度随时间衰减更快,但是单次振动持续时间衰减较慢。

Title: Calcium signal in B cell regulated by the stiffness of the substrate presenting antigen

Abstract: Poly-dimethylsiloxane (PDMS) is used to simulate antigen presentation with different stiffness substrates. Confocal microscopy is applied to collect calcium signal image data from B cells which have recognized antigen coated on substrate with different stiffness. The results show that antigens on stiffer surface induce higher frequency of occurrence of calcium oscillation compared to that on less stiffer surface, more swift amplitude attenuation, but shorter duration of single calcium oscillation.
