一种治疗胆总管结石新型药物洗脱复合金属支架的制备及表征

摘 要: 以乙二胺四乙酸(EDTA)&胆酸钠(SC)为负载药物,以聚己内酯(PCL)为药物载体,采用表面浸涂法制备一种用于胆总管(common bile duct,CBD)结石治疗的新型药物洗脱复合金属支架,并对其药物载入形式、径向受力、体外药物释放和体外溶石进行研究。研究结果表明,浸涂法制备的药物洗脱复合金属支架涂层表面呈白色、光滑平整且支架内腔畅通;光学显微镜照片、扫描电子显微镜(scanning electron microscope,SEM)照片和X射线衍射(X-ray diffraction,XRD)图谱表明,EDTA以晶体颗粒形式均匀分散在涂层内部,SC则以无定型状态存在;径向压缩力学结果显示,支架回弹性良好,径向受力不会改变支架的截面积;此外,体外药物释放实验和溶石实验表明,制备的药物洗脱复合金属支架的药物释放时间超过28 d并且在第28天结石质量减少33.3%,显示出良好的体外溶石效果。该支架有望应用于CBD结石疾病的治疗。

Abstract: A novel drug-eluting composite metal stent coated with polycaprolactone (PCL) layer in which loaded ethylene diamine tetraacetic acid (EDTA) & sodium cholate (SC) was fabricated for common bile duct (CBD) stone by the way of surface coating. And studies on EDTA&SC existing form in coating layer, radial mechanical properties, drug release and gallstone dissolution in vitrotest were carried out in this paper. Results showed that this novel drug-eluting composite metal stent prepared by surface coating possessed white smooth surface and an unobstructed lumen; as shown in the optical microscope images, scanning electron microscope (SEM) images and X-ray diffraction (XRD) spectra, uniformly dispersed EDTA particles were seen clearly in crystal state in the coating layer, but SC was in amorphous state. Radial compression mechanical properties proved perfect resilience and unchanged cross-section after removing the radical force. Besides, drug release time of this novel drug-eluting composite metal stent was over in the 28th day and the mass of gallstone reduced by 33.3% in vitro. This novel drug-eluting composite metal stent has potential application for CBD stone treatment.
