Pak2在化疗耐药卵巢浆液性癌组织和细胞中的表达及临床意义研究

摘 要: 目的:检测p21激活激酶2(p21 protein activated kinase 2,Pak2)在化疗耐药、敏感卵巢浆液性癌组织和细胞中的表达差异。方法:免疫组化、Western blotting方法检测化疗耐药、敏感卵巢浆液性癌组织中Pak2的表达情况;实时荧光定量PCR(quantitative real-time PCR,qRT-PCR)及Western blotting方法检测耐紫杉醇卵巢癌SKOV3-TR30细胞及其亲本细胞SKOV3中Pak2 mRNA及蛋白的表达。结果:免疫组化及Western blotting方法证明Pak2在化疗耐药卵巢浆液性癌组织中的表达较敏感组织显著升高,免疫组化实验证明Pak2在化疗耐药卵巢浆液性癌组织中以细胞浆表达为主。qRT-PCR表明,Pak2 mRNA在两个细胞系中表达无显著差异;Western blotting结果表明,Pak2蛋白在SKOV3-TR30细胞中较SKOV3细胞中显著高表达。结论:Pak2在化疗耐药卵巢浆液性癌组织与细胞中的表达显著升高,且与卵巢癌化疗耐药相关。

Title: Study on the expression of Pak2 in chemotherapy resistant ovarian serous cancer tissues and cells and its clinical significance

Abstract: Objective: To detect the differential expression of p21 protein activated kinase 2 (Pak2) in chemotherapy resistant and sensitive ovarian serous cancer tissues and cells. Methods: Immunohistochemistry analysis and Western blotting were applied for the detection of Pak2 in chemotherapy resistant and sensitive ovarian serous cancer tissues. Using quantitative real-time PCR (qRT-PCR) method to detect the differential expression of Pak2 in serous ovarian cancer resistant SKOV3-TR30 and sensitive SKOV3 cells. Results: Immunohistochemistry and Western blotting assay showed that the expression of Pak2 in chemotherapy resistant ovarian serous cancer tissues was significantly higher than the sensitive tissue, and mainly in the cytoplasm. By applying qRT-PCR and Western blotting, we found that there was no significant difference of Pak2 mRNA expression levels between these two cell lines. The expression of Pak2 protein was significantly over-expressed in ovarian cancer paclitaxel resistance cell line SKOV3-TR30 than in its parental cell line SKOV3. Conculsion: Pak2 expression in chemotherapy resistant ovarian serous cancer tissues and cells was significantly higher and the over expression was associated with drug resistance.
