基于Man-Fe3O4和Man-CQDs纳米探针双标记模式定量检测沙门氏菌

摘 要: 以柠檬酸为碳源,通过水热法制备得到荧光碳量子点(carbon quantum dots,CQDs),基于其表面丰富的羧基,将4-氨基苯基-α-D-吡喃甘露糖苷以酰胺键的形式修饰在其表面制备得到Man-CQDs纳米探针,并以相同的方式将4-氨基苯基-α-D-吡喃甘露糖苷修饰在商品化的Fe3O4表面,制备得到Man-Fe3O4纳米探针。在混菌溶液中,Man-Fe3O4探针选择性地识别捕获沙门氏菌,在外加磁场的作用下,分离得到Bacteria-Man-Fe3O4复合物,再利用Man-CQDs对捕获的沙门氏菌进行荧光标记,利用荧光光谱仪对双标记的沙门氏菌进行荧光测试。结果显示,沙门氏菌浓度的对数与对应荧光值IFL存在良好的线性关系,其线性方程为IFL=217.2log(c)-758.8(R2=0.957 8),线性范围为104~107 cfu•mL-1,检测限为5×103 cfu•mL-1. 本文所建立的基于磁性纳米探针的双标记模式检测沙门氏菌的方法,具有快速、高选择性和高灵敏度的优点。

Title: Quantitative detection of Salmonella by dual recognition based on Man-Fe3O4 and Man-CQDs nano-probes

Abstract: In the paper, citric acid was used as carbon source for preparing fluorescent carbon quantum dots (CQDs) by hydrothermal method. Due to the obtained CQDs rich in carboxyl group on the surface, Man-CQDs nano-probe was synthesized by immobilizing 4-aminopheny-α-D-mannopyranoside on surface of CQDs via amido bond. Man-Fe3O4 nano-probe was obtained by the same way on the commercial Fe3O4. In the mixed bacteria solution, Salmonella was selectively captured by Man-Fe3O4 nano-probe, and Bacteria-Man-Fe3O4 complexes were isolated by exerting external magnetic field. Moreover, the captured Salmonella was labeled by Man-CQDs. Salmonella labeled dual recognition was tested by fluorescence spectroscopy. The results show that there is a good linear relationship between the logarithm of the concentration of Salmonella and the corresponding fluorescence value. The linear equation is IFL=217.2log(c)-758.8 (R2=0.957 8), with the linear range of 104-107 cfu•mL-1 and the detection limit of 5×103 cfu•mL-1. A method for detection of Salmonella based on the magnetic nano-probes is developed, which has the advantages of fast, high selectivity and high sensitivity.
