针刺“调腑”对焦虑模型大鼠脑肠肽motilin表达的影响

摘 要: 目的:研究针刺“关元”、“下巨虚”穴对焦虑模型大鼠杏仁核区和十二指肠段胃动素(motilin,MTL)及其受体(motilin receptor,MTLR)水平的影响,探讨针刺“调腑”抗焦虑的部分作用机理。方法:选用清洁级SD雄性大鼠,驯养1周后随机分为空白组、模型组和针刺组。采用慢性不可预知性应激(chronic unpredictable stress,CUS)法建立焦虑模型,针刺组取“关元”、“下巨虚”穴进行干预。用高架十字迷宫(elevated plus maze,EPM)观察大鼠焦虑行为(OT%与OE%分别代表大鼠在十字迷宫开臂的活动时间与次数占总时间与总次数的百分比),酶联免疫吸附测定(enzyme-linked immunosorbent assay,ELISA)法检测大鼠脑、肠MTL和MTLR浓度以及血浆皮质酮(corticosterone,CORT)的含量。结果:1)模型组较空白组的OT%值明显降低(P<0.05)、OE%值也下降(P=0.051),针刺组较模型组的OT%值明显升高(P<0.05)、OE%值同样升高(P=0.054);2)模型组较空白组的杏仁核区MTL、十二指肠段MTLR浓度均呈下降趋势(分别为P=0.067和P=0.078),针刺组较模型组的杏仁核区MTL、十二指肠段MTLR浓度均升高,但组间未呈现统计学差异(P>0.05);3)针刺组较模型组的血浆CORT含量呈下降趋势(P=0.065)。结论:针刺“调腑”抗焦虑作用可能与其上调杏仁核区MTL及十二指肠段MTLR浓度、降低下丘脑-垂体-肾上腺(hypothalamic-pituitary-adrenal,HPA)轴活性有关联趋势。

Title: Effect of acupuncture on regulating Fu organs on expression of motilin in anxiety model rats

Abstract: Objective: Effects of acupuncturing “Guanyuan” and “Xiajuxu” on the levels of motilin (MTL) and its receptors (MTLR) in both amygdala and duodenum of rats with anxiety were investigated to explore the mechanism of anti anxiety effect of acupuncture on regulating Fu organs. Methods: Clean and male SD rats were randomly divided into control group, model group and acupuncture group after 1 weeks of domestication. In both model group and acupuncture group, anxiety model was established by chronic unpredictable stress (CUS) method. Rats in acupuncture group were treated with “Guanyuan” and “Xiajuxu”. The anxiety behavior of rats was observed by elevated plus maze (EPM) (OT% and OE% represented the percentage of time and frequency of the open arm occupying total time and total number of times of the rats in the cross maze respectively). The concentrations of MTL and MTLR in the brain and intestine and the content of plasma corticosterone (CORT) of rats were detected by enzyme-linked immunosorbent assay (ELISA) method. Results: 1) Compared with control group, OT% of model group was decreased significantly (P<0.05) and OE% was also decreased (P=0.051). Compared with model group, OT% of acupuncture group was increased significantly (P<0.05) and OE% was also increased (P=0.054). 2) Compared with control group, concentrations of MTL in the amygdala and MTLR in the duodenum of model group were decreased (P=0.067 and P=0.078). Compared with model group, concentrations of MTL in amygdala and MTLR in duodenum of acupuncture group were increased, but there was no significant difference among them (P>0.05). 3) Compared with model group, the content of plasma CORT of acupuncture group was decreased (P=0.065). Conclusion: Anti anxiety effect of acupuncture on regulating Fu organs may be related to up-regulating the concentrations of MTL in amygdala and MTLR in duodenum and down-regulating activity of hypothalamic-pituitary-adrenal (HPA) axis.
