汽爆玉米秸秆产沼气及其古菌群落解析

摘 要: 为研究汽爆玉米秸秆产沼气及其发酵过程中古菌群落的变化,以汽爆玉米秸秆为原料在总固体(total solids,TS)含量为3%的条件下进行批式中温发酵产沼气,检测发酵过程中pH值、挥发性脂肪酸(volatile fatty acids,VFAs)及沼气产量的变化,同时取样,利用变性梯度凝胶电泳(denaturing gradient gel electrophoresis,DGGE)法研究发酵过程中古菌群落的变化情况。结果表明,该系统启动迅速,料液在起始4 d内产气量较大,但随着有机酸的累积造成料液酸化,中间段的产气量减小,料液总产气量达到9 075 mL. 甲烷含量稳步上升,在第19天达到最高值,占总气体量的65.3%. DGGE分析表明,系统中古菌多样性丰富,汽爆样品与对照样品间产甲烷菌种类多样性区别明显。甲烷粒菌属(Methanocorpusculum)是贯穿整个厌氧发酵过程中最主要的产甲烷菌。其中,在厌氧发酵产气前期,发酵体系中主要以甲烷杆菌属(Methanobacterium)为主,而在厌氧发酵中后期则主要以甲烷球菌属(Methanococcus)和甲烷粒菌属为主。汽爆玉米秸秆沼气系统古菌群落结构的阐明可为秸秆沼气工程的调控提供科学数据。

Title: Biogas production using steam exploded cornstalks and its archaea community composition study

Abstract: With steam exploded cornstalks as raw material [total solids (TS) content of 3%], a mesophilic batch fermentation study was carried out to investigate archaea composition, biogas production rate, pH value and volatile fatty acids (VFAs). Archaea composition was studied using denaturing gradient gel electrophoresis (DGGE). Fermentation started rapidly, and biogas production rate of feed liquid was large within 4 d. However, the gas production decreased in the middle stage of fermentation because of the acidification of material liquid caused by VFAs accumulation and the largest gas production could reach 9 075 mL. Methane in the biogas increased stably and could be 65.3% of the total amount of gas in the nineteenth day. DGGE analysis showed that archaea diversity was rich. Archaea diversity of the steam exploded cornstalks treatment group was different from that of the control group. Methanocorpusculum dominated in the whole fermentation period; Methanobacterium dominated in the early stage of fermentation period; Methanococcus and Methanocorpusculum dominated in the middle and later periods of the fermentation. This research provides implications on the regulation of archaea composition for biogas production using steam exploded cornstalks.
