环状RNA在肝癌中的功能及作用研究

摘 要: 目的:探究环状RNA(circular RNA,circRNA)在肝癌中的功能及作用。方法:通过circinteractome数据库预测与Lin28B有结合的circRNA;由RNA结合蛋白免疫沉淀(RNA binding protein immunoprecipitation,RIP)实验及实时荧光定量PCR(quantitative real-time PCR,qRT-PCR)确定Lin28B与所选基因的circRNA结合情况;由circRNA慢病毒表达载体pLO-ciR构建circRNA过表达载体;qRT-PCR检测肝癌细胞系中circRNA过表达水平;Western blotting检测circRNA过表达肝癌细胞系中Lin28B蛋白表达水平。结果:circRNA与RNA结合蛋白Lin28B存在相互结合;成功构建circRNA过表达载体;circRNA在肝癌细胞系中有显著的过表达效果;circRNA过表达的肝癌细胞系中Lin28B蛋白表达下降。结论:circRNA在肝癌细胞系中发挥调控Lin28B表达的作用。

Title: Role and function of circular RNA in hepatocellular carcinoma

Abstract: Objective: To explore the function and mechanism of circular RNA (circRNA) in hepatocellular carcinoma. Methods: The interaction of Lin28B and circRNA was predicted by circinteractome database. The circRNA binding of Lin28B to the selected gene was determined by RNA binding protein immunoprecipitation (RIP) and quantitative real-time PCR (qRT-PCR). circRNA overexpression vector was constructed by circRNA lentiviral expression vector pLO-ciR. qRT-PCR was used to detect the overexpression of circRNA in hepatocellular carcinoma cell lines. The expression of Lin28B protein in hepatocellular carcinoma cell line was detected by Western blotting. Results: Lin28B, the RNA binding protein, interacted with circRNA. circRNA overexpression vectors were successfully constructed. circRNA were successfully overexpressed in hepatocellular carcinoma cell lines. The expression of Lin28B protein in the circRNA overexpressing hepatocellular carcinoma cell lines was decreased. Conclusion: circRNA plays an important role in the regulation of Lin28B expression in hepatocarcinoma cell lines.
