不同时程铅暴露对海马神经元AMPA受体表达的影响

摘 要: 利用培养的原代海马神经元体系,采用电生理、免疫细胞化学和分子生物学等方法,研究不同时程铅暴露对突触基本传递和α-氨基-3-羟基-5-甲基-4-异恶唑丙酸(α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid,AMPA)受体表达的影响。结果发现,神经元分别铅暴露5 min、15 min、4 h和24 h,微小的兴奋性突触后电流(miniature excitatory post-synaptic currents,mEPSC)的幅度和频率均无明显变化;在铅暴露5 min、15 min和4 h后,AMPA受体亚基GluR1和GluR2总体蛋白表达量没有明显变化。但是在铅暴露15 min后,免疫细胞化学和蛋白质免疫印迹实验发现兴奋性突触后膜上GluR1蛋白量明显升高。综上,急性铅暴露15 min能引起GluR1突触膜转运的效能。

Title: Effects of lead exposure for different time on AMPA receptor expression of hippocampal neurons

Abstract: This research used primary cultured hippocampal neurons and adopted electrophysiological, immunocytochemistry and molecular biology methods to investigate the effects of different time lead exposure on synaptic basic transmission and α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptor expression. The results found that amplitude and frequency of miniature excitatory post-synaptic currents (mEPSC) had no significant changes after neurons lead exposure for 5 min, 15 min, 4 h and 24 h, respectively. Total protein expression of AMPA receptor subunits GluR1 and GluR2 had no obvious changes after lead exposure for 5 min, 15 min and 4 h. But the GluR1 protein expression of excitatory post-synaptic membrane increased significantly after lead exposure for 15 min. In conclusion, 15 min of acute lead exposure could cause the trafficking efficiency of GluR1 synaptic membrane.
