手机辐射对睾丸中Bcl-2和CYP11A1表达的影响

摘 要: 为探讨手机辐射对雄性小鼠睾丸的影响,选用60 只30 日龄健康雄性昆明小鼠为研究对象。在室温条件下常规饲养1 周以适应环境,之后随机分为4 组,分别为对照组(0 h/d)和手机辐射1 h/d 组(低辐射组)、2 h/d 组与3 h/d 组(高辐射组),每组15只,第31 天处死,取其睾丸组织,进行各项试验检测。苏木精-伊红(hematoxylin-eosin,HE)染色观察睾丸组织病理学变化;免疫组织化学法和Western blotting 检测Bcl-2 和Cyp11a1 蛋白的分布和表达量;反转录PCR(reverse transcription-PCR,RT-PCR)法检测睾丸组织中Bcl-2 mRNA和Cyp11a1 mRNA 的表达水平。研究结果表明:通过HE 染色观察小鼠睾丸组织结构,实验组与对照组相比无明显病理变化;免疫组织化学法观察到Bcl-2 蛋白的表达随辐射时长的增加而降低。同时发现,与对照组相比,高辐射组Cyp11a1 蛋白的表达量也降低;且mRNA 水平符合蛋白表达的趋势,Western blotting 的验证结果也是如此。综上,手机辐射虽然没有引起睾丸组织结构明显的改变,但其通过影响相关蛋白的表达而影响睾丸的生理机能。

Title: Effect of mobile phone radiation on the expression of Bcl-2 and Cyp11a1 in testis

Abstract: To study how mobile phone radiation influences the testis of male mice, 60 30-day-old male healthy Kunming mice were chosen in the experiment. After being regularly fed under indoor temperature for one week, the mice had adapted to the environment, and were divided into 4 groups randomly, as mobile phone radiation for 1 h/d, 2 h/d, 3 h/d and control (0 h/d) groups. Each group had 15 mice and they were put to death at 31st day. Then the testis was taken from each mouse for test. Through hematoxylin-eosin (HE) staining, the pathological changes of the testis tissue were observed. The disposition and expression of Bcl-2 and Cyp11a1 proteins were investigated using immunohistochemistry and Western blotting, and the expressions of Bcl-2 mRNA and Cyp11a1 mRNA in the testis were tested by reverse transeription-PCR (RT-PCR). The results showed that there was no obvious pathological change between experiment and control groups by HE staining. The expression of Bcl-2 protein decreased when the radiation time increased by immunohistochemistry. The expression of Cyp11a1 protein in 3 h/d group also decreased compared to the control group. Moreover, the level of mRNA was in accordance with the trend of protein expression, which was proved by Western blotting. In conclusion, the mobile phone radiation could not cause obvious structural change of testis, but could make change of protein expression to influence the physiological function of testis.
