人参皂甙Rb1通过增加AQP4表达治疗SCII的研究

摘 要: 目的:探讨人参皂甙Rb1对脊髓缺血再灌注损伤(spinal cord ischemia-reperfusion injury,SCII)大鼠水通道蛋白4(aquaporin 4,AQP4)表达的影响。方法:建立大鼠SCII模型,96只SD大鼠随机分为4组,每组24只,分别为空白对照组、假手术组、安慰剂组和人参皂甙Rb1治疗组,治疗组连续7 d给予腹腔注射人参皂甙Rb1 10 mg•kg-1•d-1. 各组分别在灌注后的1,3,5,7 d四个时间点处死6只大鼠。Basso Beattie Bresnahan(BBB)评分系统评价神经功能,苏木精-伊红(hematoxylin-eosin,HE)染色检测脊髓组织结构和细胞形态变化,TUNEL法检测细胞凋亡情况。分别采用免疫荧光法和real-time PCR方法检测AQP4蛋白和mRNA表达变化。结果:与安慰剂组比较,人参皂甙Rb1治疗可显著增加大鼠BBB评分,改善神经细胞形态,减少细胞凋亡,显著增加SCII模型的AQP4蛋白和mRNA表达,并且在人参皂甙Rb1干预7 d后AQP4蛋白和mRNA表达水平恢复至正常水平,与空白对照组比较无显著差异(P>0.05)。结论:人参皂甙Rb1对大鼠SCII具有治疗效果,其作用机制可能是上调AQP4表达,减少神经水肿及细胞凋亡。

Title: Treatment SCII with ginsenoside Rb1 by increasing AQP4 expression

Abstract: Objective: To investigate the effect of ginsenoside Rb1 on aquaporin 4 (AQP4) expression in spinal cord ischemia-reperfusion injury (SCII) rats. Methods: The SCII model of rats was established. 96 SD rats were randomly divided into four groups with 24 rats in each group. The groups were blank control group, sham operation group, placebo group and ginsenoside Rb1-treated group. In the ginsenoside Rb1-treated group, intraperitoneal injection was given with ginsenoside Rb1 10 mg•kg-1•d-1 for 7 d. At 1, 3, 5, 7 d after reperfusion, 6 rats of each group were then killed, and the following studies were performed. The scores of spinal cord injury were determined by Basso Beattie Bresnahan (BBB) system, the spinal cord tissue structure and cell morphology were analyzed by hematoxylin-eosin (HE) staining, and the apoptosis rate was measured by TUNEL. The AQP4 protein and mRNA expressions were detected with immunofluorescence and real-time PCR. Results: Comparing with the placebo group, ginsenoside Rb1 treatment could significantly increase the BBB scores of rats, improve the morphologies of nerve cells, decrease cell apoptosis, and increase the AQP4 protein and mRNA expressions in SCII rat model. After treatment with the ginsenoside Rb1 intervention for 7 d, the expression levels of AQP4 protein and mRNA were returned to normal levels, and there was no significant difference when compared to the blank control group (P>0.05). Conclusion: Ginsenoside Rb1 had treatment effects on SCII rats. The possible mechanism may be involved with up-regulation of AQP4 expression, and reduction of nerve edema and cell apoptosis.
