黄芪总苷对高糖所致人肾小球系膜细胞损伤的保护作用及机制研究

摘 要: 目的:研究黄芪总苷(astragalosides,AST)对高糖诱导的人肾小球系膜细胞(human mesangial cell,HMC)损伤的保护作用及其机制。方法:将HMC作为目标细胞,分别设立正常对照组(NG)、甘露醇渗透压对照组(MA)、高糖组(HG)、Tempol组(100 μmol/L)及不同剂量AST组(50,100,200 mg/L)。干预24 h后,噻唑蓝(MTT)法检测AST(0~400 mg/L)对高糖培养HMC细胞增殖与细胞活力的影响;流式细胞术检测各组HMC中活性氧(reactive oxygen species,ROS)含量;Western blotting法检测各组HMC中TRPC6、NOX4蛋白表达情况;生物素双抗体夹心酶联免疫吸附测定(enzyme linked immunosorbent assay,ELISA)法分析各组HMC上清液中纤维连接素(fibronectin,FN)、IV型胶原(Col IV)含量。结果:在0~400 mg/L浓度范围内,AST可呈浓度依赖性地抑制高糖培养HMC细胞增殖。与HG组相比,AST可在一定程度上降低HMC上清液中FN、Col IV含量,减少HMC中ROS含量;显著提高TRPC6表达水平,上调HMC中TRPC6蛋白表达水平,下调NOX4蛋白表达水平。结论:AST对高糖诱导的HMC的损伤具有保护作用,其机制可能与AST提高HMC抗氧化能力、降低FN和Col IV含量、抑制NOX4高表达以及提高TRPC6表达水平有关。

Title: Protective effects of astragalosides on high glucose induced human mesangial cell injury and its mechanisms

Abstract: Objective: To research the protective effects of astragalosides (AST) on high glucose induced human mesangial cell (HMC) injury and its potential mechanisms. Methods: HMC was taken as target cell. The HMCs were divided into seven groups and treated as follows: normal group (NG), mannitol osmotic pressure control group (MA), high glucose group (HG), Tempol group (100 μmol/L) and AST group with different doses (50, 100, 200 μmol/L). After treating for 24 h, the effects of AST (0-400 mg/L) on cell proliferation and cell viability of HMC in high glucose culture were detected by MTT assay. The reactive oxygen species (ROS) production in HMC was measured by flow cytometry. Western blotting was used to test the expression of TRPC6 and NOX4 proteins in HMC. The contents of fibronectin (FN) and Col IV in the supernatant were detected by biotin double antibody sandwich enzyme linked immunosorbent assay (ELISA). Results: At the range of 0-400 mg/L, the cell proliferation inhibiting caused by AST showed dose-dependent in high glucose incubated HMC. Compared with HG group, AST group could decrease the contents of FN and Col IV in the supernatant, inhibite the contents of ROS in HMC, significantly increase TRPC6 expression, increase TRPC6 protein expression in HMC and decrease NOX4 protein expression. Conclusion: There is protective effect of AST on high glucose incubated HMC injury. The potential mechanisms may include the enhancement of anti-oxidative ability of HMC, reduction of the contents of FN and Col IV, inhibition in the high expression of NOX4 and promotion of the TRPC6 expression by AST.
