黄芪甲苷预处理MSCs移植对心梗后心衰大鼠心功能的改善作用

摘 要: 目的:比较黄芪甲苷预处理骨髓间充质干细胞(mesenchymal stem cells,MSCs)前后,干细胞移植对心梗后心衰大鼠心功能的影响。方法:通过左冠状动脉前降支(left anterior descending coronary artery,LAD)结扎构建心梗后心衰大鼠模型,设置假手术组、模型组、MSCs移植组和黄芪甲苷预处理MSCs移植组,比较移植4周后各组大鼠左心室射血分数(left ventricular ejection fraction,LVEF)的差异,并对心脏切片进行α-actinin免疫荧光染色,标记心肌细胞,观察MSCs存活情况。结果:与假手术组相比,模型组的LVEF显著下降(P<0.001),MSCs移植组、黄芪甲苷预处理MSCs移植组的LVEF均显著提高(P<0.01,P<0.001),其中黄芪甲苷预处理MSCs移植组改善LVEF的效果较MSCs组有上升趋势(P>0.05)。免疫荧光染色结果显示,4周后,MSCs在黄芪甲苷预处理组存活更多。结论:经黄芪甲苷预处理后,MSCs移植疗法或能更好地改善心衰大鼠的心功能,其可能是通过提高MSCs存活率作用的。

Title: Improvement of cardiac function via transplantation of astragaloside IV-pretreated MSCs in post-myocardial infarction heart failure rat models

Abstract: Objective: To compare the effect of improvement on cardiac function between mesenchymal stem cells (MSCs) and astragaloside IV-pretreated MSCs (AST-MSCs) transplantation in post-myocardial infarction heart failure rat models. Methods: The post-myocardial infarction heart failure rat models were established by left anterior descending coronary artery (LAD) ligation. The sham surgery group, model group, MSCs transplantation group, and AST-MSCs transplantation group were built, and the differences of left ventricular ejection fraction (LVEF) among these groups were compared 4 weeks after transplantation. Meanwhile the myocardium of heart slice was marked by α-actinin immunofluorescence staining, and the survival of MSCs was observed. Results: Compared to sham surgery group, LVEF of model group showed significantly decrease (P<0.001). Both MSCs and AST-MSCs transplantation groups significantly improved LVEF (P<0.01, P<0.001), AST-MSCs transplantation group showed a trend of better up regulation of LVEF (P>0.05). Immunofluoresecent staining results indicated that, 4 weeks after transplantation, there were more MSCs survival in AST-MSCs transplantation group. Conclusion: AST-MSCs transplantation may better improve the cardiac function of post-myocardial infarction heart failure rat models, and the mechanism might be the promotion of MSCs survival.
