复方丹参有效组分及其组合物对MIF诱导人神经母细胞瘤细胞SH-SY5Y细胞凋亡的作用

摘 要: 为探究复方丹参(Fufang Danshen,FFDS)有效成分及其组合物对巨噬细胞移动抑制因子(macrophage migration inhibitory factor,MIF)诱导人神经母细胞瘤细胞SH-SY5Y细胞凋亡的作用机制,采用噻唑蓝(MTT)法检测细胞生长情况;利用流式细胞仪技术检测细胞凋亡;通过Western blotting法检测目标蛋白的表达。研究结果表明,MIF表现出与Aβ1-42相似的神经毒性,能够促进SH-SY5Y细胞的凋亡。MIF可通过诱导SH-SY5Y细胞中Bad的表达量增加来引起细胞凋亡。而复方丹参组合物、丹参酮IIA(tanshinone IIA,Tan IIA)和三七皂苷R1(notoginsenoside R1,NG-R1)能够显著抑制MIF诱导的细胞凋亡,并能明显减少Bad的表达量。进而推测,复方丹参组合物可能通过下调Bad的表达而发挥神经保护作用。

Title: Role of the effective components of Fufang Danshen and their composition in MIF-induced apoptosis in human neuroblastoma cells SH-SY5Y

Abstract: The objective of this study are explored the mechanism of effective components of Fufang Danshen (FFDS or Danshen compound) and their composition (FFDS’s composition) on macrophage migration inhibitory factor (MIF)-induced apoptosis in human neuroblastoma cells SH-SY5Y. Here, cell growth, apoptosis and target protein expression are detected by MTT assay, flow cytometry and Western blotting, respectively. The results show as follows: MIF presents neurotoxicity similar to Aβ1-42 and can promote apoptosis of SH-SY5Y cells. MIF can induce the high expression of Bad in SH-SY5Y cells and then trigger apoptosis. FFDS’s composition, tanshinone IIA (Tan IIA) and notoginsenoside R1 (NG-R1) can obviously inhibit apoptosis and reduce the expression of Bad in MIF-induced SH-SY5Y cells. Therefore, we speculate that FFDS’s composition might play a role in the protection of nerve through down-regulating the expression of Bad in neurodegenerative diseases.
