长链非编码RNA AK124173通过NF-κB通路应答脂多糖刺激

| 摘 要： | 目的：研究长链非编码RNA（long non-coding RNA，lncRNA）AK124173通过何种途径应答脂多糖（lipopolysaccharide，LPS）刺激，以及该基因表达变化对其他炎症因子的影响。方法：筛选LPS刺激能上调幼稚单核细胞（U937）基因表达的lncRNAs，进而实验确认筛选的lncRNA不编码蛋白。同时通过信号通路抑制剂处理细胞，分析AK124173基因转录调控机制，并通过荧光素酶实验验证。通过siRNA技术和过表达分析AK124173基因的表达变化对其他炎症因子的影响。结果：在幼稚单核细胞中，AK124173基因能被LPS（200 ng/mL）刺激上调表达10倍，AK124173基因不具备开放阅读框结构，属于非编码RNA范畴。NF-κB通路抑制剂SC-514能下调AK124173基因的mRNA表达。过表达AK124173能导致IL8基因的启动子活性增强，但无论是过表达还是siRNA AK124173对旁边TNFAIP3（A20）基因的影响均不明显。结论：AK124173是一个lncRNA，其转录受到NF-κB信号通路调控，但该基因的表达变化并不影响临近基因A20的表达。 |

| Abstract： | Objective: To investigate the pathway of long non-coding RNA (lncRNA) AK124173 respond to lipopolysaccharide (LPS) stimulation and the effects of this gene expression changes on other inflammatory factors. Methods: lncRNAs of gene expression of U937 cell lines upregulated by LPS stimulation were screened, and then the confirmatory screening of lncRNA did not code protein. At the same time, the transcriptional regulation mechanism of AK124173 gene was analyzed by signaling pathway inhibitor, and it was verified by fluorescein experiment. The effect of AK124173 gene expression changes on other inflammatory factors was analyzed by siRNA knockdown and gene overexpression. Results: In naive monocytes cells, AK124173 gene could be upregulated for 10-folds by LPS (200 ng/mL) stimulation. AK124173 gene had no open reading frame, and belonged to the category of non-coding RNA. The NF-κB pathway inhibitor SC-514 could downregulate the mRNA expression of AK124173 gene. Overexpression of AK124173 could enhance the promoter activity of IL8, however, neither overexpression nor siRNA AK124173 had significant effects on the adjacent TNFAIP3 (A20) gene. Conclusion: AK124173 is a kind of lncRNA whose transcription is regulated by the NF-κB signaling pathway, but the expression of the gene does not affect the expression of the adjacent gene A20. |
