原代HIV-1 B’亚型包膜gp120三聚体蛋白抗原性及免疫原性研究

摘 要: 目的:研究I型人类免疫缺陷病毒(human immunodeficiency virus type 1,HIV-1)B’亚型广谱中和者(CNHLJBF06044)体内病毒包膜gp120三聚体蛋白(gp120T)的抗原性和免疫原性。方法:通过假病毒中和试验分析06044包膜假病毒与B’亚型感染者血浆的中和活性;通过酶联免疫吸附测定(enzyme linked immunosorbent assay,ELISA)试验分析06044 gp120T与已知广谱中和抗体(broadly neutralizing antibodies,BNAbs)的结合活性;06044 gp120T免疫BALB/c小鼠,通过ELISA和中和试验分析其免疫血清抗原特异性结合抗体及中和抗体水平。结果:1)含有06044包膜假病毒可被56%(14/25)的HIV-1 B’亚型感染者血浆中和,半数抑制量(50% inhibitory dose,ID50)值的几何平均滴度(geometric mean titer,GMT)为1:1 084;2)06044 gp120T表面中和抗体表位暴露良好,可与多个BNAbs结合;3)06044 gp120T免疫小鼠后,产生交叉反应性结合抗体和中和抗体。结论:HIV-1 06044 gp120T具有良好的抗原性,在小鼠试验中具有良好的免疫原性。

Title: Antigenicity and immunogenicity of the envelope gp120 trimer derived from a primary HIV-1 subtype B’ isolate

Abstract: Objective: To investigate the antigenicity and immunogenicity of human immunodeficiency virus type 1 (HIV-1) subtype B’ envelope (Env) gp120 trimer (gp120T) derived from a broad neutralizer (CNHLJBF06044). Methods: The neutralizing activity of 06044 Env-pseudovirus and subtype B’ infected plasma was analyzed by pseudovirus neutralization assay. Binding activities of 06044 gp120T to known broadly neutralizing antibodies (BNAbs) were detected by enzyme linked immunosorbent assay (ELISA). Binding and neutralizing antibody responses in 06044 gp120T-immunized mice BALB/c were measured by ELISA and neutralization assays, respectively. Results: 1) 06044 Env-pseudovirus could be neutralized by 56% (14/25) of HIV-1 B’ subtype infected plasma with the 50% inhibitory dose (ID50) geometric mean titer (GMT) of 1:1 084. 2) 06044 gp120T had well-exposed epitopes on its surface and could bind to multiple BNAbs. 3) After immunization with 06044 gp120T, cross-reactive binding and neutralizing antibody responses were induced in mouse sera. Conclusion: HIV-1 06044 gp120T had well antigenicity and strong immunogenicity in mice.
