离子液体溶解法制备高强度血管用胶原膜材料

摘 要: 采用离子液体溶解法制备高强度血管用膜材料。以离子液体(1-丁基-3-甲基咪唑氯盐)溶解胶原蛋白,并进行肝素化修饰得到高强度血管用胶原膜材料。材料力学性能测试结果表明,膜材料具有良好的弹性和抗拉强度。扫描电子显微镜(scanning electron microscope,SEM)观察结果显示膜材料表面为多孔网状结构。细胞活性实验表明,材料具有明显的促细胞增殖作用。血液相容性测试结果表明,随着肝素含量的增加,肝素化胶原膜材料活化部分凝血活酶时间(activated partial thromboplastin time,APTT)显著延长,血小板粘附量降低,说明肝素极大地提高了膜材料的血液相容性。综上所述,离子液体溶解法制备的肝素化胶原膜材料在组织工程领域具有广阔的应用前景。

Title: Preparation of high strength collagen film for blood vessels by ionic luquid dissolving method

Abstract: Vascular membrane materials with high strength were prepared through ionic liquid dissolving method. Ionic liquid (1-butyl-3-methylimidazolium chloride) was used to dissolve collagen, heparin modification was used to obtain high strength collagen film for blood vessels. The material mechanics performance test result showed that the as-prepared heparin-collagen film had excellent elasticity and tensile strength. Scanning electron microscope (SEM) observations suggested that the prepared film surface had a porous structure. Cell viability assay indicated that the film significantly facilitated the cell proliferation. Blood compatibility test showed that, with the increase of heparin content, activated partial thromboplastin time (APTT) of heparin membrane materials significantly increased while the number of platelet adhered on the film decreased. It indicated that heparin effectively improved the blood compatibility of the film. In summary, heparin-collagen film prepared by ionic liquid dissolving method had strong potential in the field of tissue engineering.
